姜黄素对IL-1β诱导视网膜色素上皮细胞分泌IL-8和sICAM-1的影响 被引量:4

摘要 目的:研究姜黄素对IL-1β诱导RPE细胞分泌IL-8和sICAM-1的影响。方法:RPE细胞经含4mL/LFBS的DMEM/F12同步后分为四组:①rhIL-1β干预组;②rhIL-1β+姜黄素组,姜黄素分5mg/L和10mg/L两个浓度;③姜黄素组,只分别加入5mg/L和10mg/L姜黄素;④空白对照组(4mL/LFBS+DMEM/F-12)。所有样本干预24h后收集培养上清液和该孔细胞蛋白裂解液。ELISA法检测rhIL-1β(5μg/L)和姜黄素诱导hfRPE细胞分泌IL-8和sICAM-1的量。结果:rhIL-1β(5μg/L)组RPE细胞分泌的IL-8和sICAM-1分别比空白对照组升高了20倍和8倍,加入姜黄素能显著抑制其分泌。5mg/L姜黄素使IL-8和sICAM-1含量分别降低了36.88%和14.90%,10mg/L则分别达到55.73%和41.78%。姜黄素本身不刺激hfRPE细胞分泌IL-8和sICAM-1。结论:姜黄素可显著抑制IL-1β刺激的RPE细胞分泌炎前因子IL-8和sICAM-1,有可能防止PVR的发生。 AIM:To investigate the effect of curcumin on rhIL-1β -induced IL-8 and sl CAM-1 secretion in RPE cells. METHODS:Synchronized RPE cells in vitro were treated for 24 hours with medium alone or medium containing rhlL-1β (Sμg/L), curcumin (5 or 10mg/L), combination of rhIL-1β (Sμg/L) and curcumin (5 or 10mg/L), respectively. ELISA was employed to measure the secreted I L-8 and slCAM-1 in cell-culture supernatants. Bradford protein assay was performed to determine the protein concentration of the cell lysate. Secreted I L-8 and sl CAM-1 protein (ng/g) were standardized by expressing the data to cellular protein in each lysate. RESULTS:Compared with negative control, rhlL-1β -induced I Lo8 and slCAM-1 expression increased 20-fold and 8-fold, respectively. Curcumin partially blocked the inductive effeds. The protein secretion of IL-8 and slCAM-1 were reduced 36. 88% and 14.90% respectively by curcumin at 5mg/L, 55.73% and 41.78% respectively by that at 10mg/L. Curcumin itself did not induce IL-8 and slCAM-1 protein secretion.CONCLUSION: Curcumin can significantly inhibit overexpression of IL-1β-induced IL-8 and slCAM-1 in RPE cells, thus it maybe a potential candidate in PVR pharmacotherapy.
