摘要 目的探讨氧化低密度脂蛋白(OX-LDL)对小鼠视网膜色素上皮细胞(RPE)细胞的炎症诱导及其与Toll样受体4(TLR4)的相关性。方法采用酶消化法进行野生型及TLR4基因敲除(TLR4-/-)的C57小鼠RPE细胞的体外原代培养,通过免疫化学染色对培养的RPE细胞进行鉴定。分别给予小鼠RPE细胞10-100 mg/ml原生低密度脂蛋白(n-LDL)或OX-LDL刺激24 h。采用ELISA法检测各组细胞培养上清液中白介素-6(IL-6)、白介素-1β(IL-1β)的含量,Western Blot及RT-PCR法检测各组细胞中TLR4的蛋白质及mRNA含量。结果 50、100 mg/ml OX-LDL组野生型小鼠RPE细胞IL-6、IL-1β表达增高,伴随TLR4蛋白质及mRNA表达升高,其差异与对照组及nLDL组相比有统计学意义。N-LDL与OX-LDL组TLR4-/-小鼠RPE细胞未见IL-6、IL-1β表达明显升高。结论OX-LDL可诱导小鼠RPE细胞产生炎症反应,其效应机制与TLR4激活相关。 Objective To study the inflammatory secretion of mouse retinal pigment epithelium( RPE) induced by oxidized low-density lipoprotein( OX-LDL) and the potential role of Toll-like receptor 4( TLR4) in this process. Methods RPE cells of wild and gene knock-out( TLR4-/-) C57 mice were primary cultured in vitro with enzyme digestion. The cultured RPE cells were identified with immunocytochemical staining. RPE cells were incubated with 10- 100 mg / m L native-LDL( n-LDL) or OX-LDL for 24 h. IL-6 and IL-1β protein in the supernatant was measured by enzyme-linked immune sorbent assay( ELISA). Western blot analysis and reverse transcription polymerase chain reaction( RT-PCR) were used to detect TLR4 protein and mRNA levels in RPE cells. Results 50 and 100 mg / m L OX-LDL treatment increased IL-6 and IL-1β secretion in wild mouse RPE cells,with TLR4 mRNA and protein expression activated. There was no significant IL-6 and IL-1β secretion increase in TLR4-/- mouse RPE cells. Conclusion Through TLR4 upregulation,OX-LDL induces inflammation promotion in cultured mouse RPE cells.
