摘要
采用发酵法生产得到的富含谷胱甘肽的产朊假丝酵母提取液,含有蛋白质、多糖、核酸等杂质,严重影响到提取高纯度的谷胱甘肽。本文研究了应用PES超滤膜对富含谷胱甘肽的产朊假丝酵母提取液进行超滤的情况,同时分析了膜压差△P、温度t、pH、料液浓度等因素对超滤的影响,得到了较好的工艺操作条件:使用截留分子量为5000的PES超滤膜,△P=0.2MPa、t=24℃、pH=1~3,最终得到的滤液中的谷胱甘肽回收率达到80%,蛋白质脱除率达到38%。
There were many impurities such as protein, amylase, nucleic acid that seriously affected the purity of glutathione in the Candida utilis WSH 02-08 cell solution obtained by fermentation. This paper studied the removal of proteins in the Candida utilis WSH 02 -08 cell solution by applying rolling membrane (PES) in the ultrafiltration device, Through analyzing the effects of some parameters such as the pressure (AP), the temperature (t) , the concentration, the value of pH on ultrafiltration process, the better operational conditions was obtained . As for the rolling membrane of which the retentate molecular weight is 5000, the optimum technology conditions were AP=0.2MPa, t=24℃, pH=l-3. Eventually, the recovery rate of GSH in the Candida utilis WSH 02-08 cell solution after ultrafiltration was 80%, and the removal rate of proteins was up to 38%.
