摘要
目的探讨参附注射液(Shenfuinjection,SF)对大鼠移植肝脏缺血再灌注损伤的保护作用及机制。方法采用雄性SpragueDawley(SD)大鼠同种异体原位肝移植模型,60只SD大鼠随机平均分成对照组和SF处理组,移植肝脏再灌注3、6、24h取血及肝脏组织检测。结果SF组血清超氧化物歧化酶(SOD)和一氧化氮(NO)水平明显高于对照组(P<0·05),丙氨酸转氨酶(ALT)、天冬氨酸转氨酶(AST)、乳酸脱氢酶(LDH)、透明质酸(HA)、丙二醛(MDA)、肿瘤坏死因子α(TNF-α)、白介素1(IL-1)、内皮素1(ET-1)水平和肝脏细胞凋亡指数明显低于对照组(P<0·05),肝脏组织形态学改变也轻于对照组。结论参附注射液对移植肝脏缺血再灌注损伤具有防治作用,可能的机制包括抑制枯否细胞激活和氧自由基产生,改善微循环,减少细胞凋亡。
Objective To investigate the effect of Shenfu injection on ischemia reperfusion injury of rat liver graft. Methods Male SD rats were used as isogeneic orthotopic liver transplantation. Sixty rats were randomly divided into two groups (30 in each). The recipient received Shenfu injection intravenously immediately before the removal of the liver in SF group and normal saline of the same volume in control group. At 3,6 and 24 h after the reperfusion blood and hepatic tissue were sampled for examination. Results The levels of superoxide dismutase (SOD) and nitric oxide (NO) in SF group significantly increased compared with those in control group ( P 〈 0. 05). The levels of serum liver enzymes, hyaluronic acid (HA) ,malondialdehyde (MDA) ,TNF-α,IL-1 ,endothelin -1 ( ET-1 ) and liver cell apoptosis index in SF group were lower than in control group ( P 〈 0. 05 ). Microscopic examination revealed that the morphological changes of hepatic tissue in control group were more severe than in SF group. Conclusions Shenfu injection has protective effect on ischemia reperfusion injury of rat liver graft. It inhibits the production of oxygen free radical and the activation of Kupffer cells, decreases apoptosis of liver cell and improves microcirculation.
