摘要
刺五加(Acanthopanaxsenticosus)胚性愈伤组织经根癌农杆菌(Agrobacteriumtumefaciens)C58C1(pBZ6111)感染后,共培养48~72h,再转移到含氯霉素50mg/L的MS选择培养基(附加2,4-D0.5mg/L,NAA0.2mg/l)上继代培养50~60d,筛选得到氯霉素抗性愈伤组织.该愈伤组织能在无激素的MS培养基上增殖,并检测到胭脂碱合成酶活性,表明是转化愈伤组织.转化愈伤组织可诱导形成胚状体,但胚状体分化形成的苗不能生根,可能是由于转化细胞内源细胞分裂素含量过高,抑制了根的分化.
in the report, we studied transformation of Acanthopanax senticosuscalli. The calli were infected with Agrobacterium tumefaciens strain C58C1 (pBZ6111) and cultured on MS sclective medium containing chloramphenical 50 mg/L,2, 4-D 0.5 mg/L and NAA 0. 2 mg/L to select transformants.Thechloramphenical-resistant calli were obtained when the Agrobacterium-infected calliwere subcultured on the MS selective medium for 50-60 days. The transformedcalli could grow on MS medium without phytohormone, and confirmed by analysisof nopaline synthetase(NOS)activity. Some embryoids from the transformed calligerminated and developed into shoots but non-rooted.
