刺五加离体培养中胚状体的诱导 被引量:10

摘要 目的:研究2 ,4 - D及BA两种植物激素相互配比以及不同的外植体在刺五加胚状体诱导中的作用。方法:采用组织培养方法。以MS为基本培养基,附加2 ,4 - D及BA。以刺五加种胚及无菌苗的子叶、叶柄、胚轴、胚根等不同外植体为材料。定期观察记录胚状体的发生及生长情况。结果:子叶及胚轴较易产生胚状体(1~2个月)。诱导胚状体的最佳培养基为MS+2 ,4 - D 0 .5 mg/ L。结论:不同的外植体及激素水平诱导胚状体的效果不同,2 ,4 - D起主导作用;培养条件以黑暗比在光照下效果好。 Objective:To study the effects of di ff erent combinations from 2,4-D and BA and different explant on the inducement of embryoids in A.senticosus.Methods:Swollen cotyledon,leafstalk,embryo axis and radicl e of test tube seedling and the embryos of A.senticosus were planted on MS m edium supplemented with 2,4-D and BA. Formation and development of embryoids wer e observed and determined.Results:Embryoid is easy to be produced on swollen cotyled on and embryo axis (1 to 2 months). The optimal medium of producing embryoids is MS+2,4-D0.5 mg/L.Conclusion:Different combinations of 2,4-D and BA and diff erent explant have different effects on the inducement of embryoids of A.Sentico sus,while 2,4-D is the key factor. Meamwhile, the effect of inducement is much b etter in dark than in light.
