摘要
用人食管癌Eca109、人宫颈癌HeLa细胞株和艾氏腹水癌(EAC)小鼠检测了马氏钳蝎 蝎毒(SVC)体外、体内的抗肿瘤作用。结果显示,SVC的浓度为0. 017 mg/L,处理Eca 109细胞24、 48、72h后,细胞生长抑制率分别为 35. 6%、39. 5%、36, 9%;MTT显色法表明,SVC浓度为0. 017、 0.034、0.085 mg/L时,对Eca109细胞的毒性作用分别达 63%、56%、59%。 SVC对体外培养的 HeLa细胞作用不明显。 SVC腹腔注射于EAC小鼠,其剂量为每kg体重 0. 1、0. 3 mg,连续给药 10 d,EAC小鼠的生命延长率分别为 52.0%、54.4%,差异显著(P<0. 05);在停药第 10 d时,EAC小 鼠的体重抑制率亦有明显差异。结果证明:SVC对Eca 109细胞和EAC小鼠有抑制作用,对HeLa 细胞作用不明显。
The human cancer cell lines,HeLas and Eca 109 cells,were incubated with Scorpion venom crude (SVC). The mice with Ehrlich ascites carcinoma (EAC) were injected with SVC. The antineoplastic effects of SVC were studied in vitro and in vivo. The results showed that while Ecal09 cells were incubated with SVC for 24, 48,and 72h,the rates of inhibition were 35. 6%, 39. 5%, 36. 9%, respectively. The concentration of SVC used was 0. 017 mg/L. The results of MTT method indicated that when the concentration of SVC were 0. 017 mg/L, 0. 034 mg/L,and 0. 085 mg/L, the cytotoxicity were 63%, 56% and 59%, respectively. But SVC showed no apparent effect on HeLas. The EAC mice were injected with SVC 0.1 mg/kg and 0. 3 mg/kg for 10 d,and the rates of prolongated survival time of EAC mice were 52. 0% and 54.4% respectively. The difference were significant. When the injection of SVC was stopped 10 d later,the difference in the inhibition rates of weight was also significant. The results attest that SVC has inhibitory effects on Eca 109 cells and EAC mice,while SVC has no apparent effects on HeLa cells.
