摘要
目的探讨p57kip2、cyclinD1及cyclinE蛋白在乳腺癌发生、发展中作用。方法用免疫组化SP法检测64例乳腺浸润性导管癌(invasiveductalcarcinoma,IDC)、15例乳腺导管原位癌(ductalcarcinomainsitu,DCIS)和15例癌旁正常乳腺组织中p57kip2、cyclinD1和cyclinE蛋白的表达情况。结果p57kip2、cyclinD1和cyclinE蛋白在IDC的阳性率与在乳腺不同组织之间相比,cyclinD1、cyclinE蛋白在DCIS的阳性率与癌旁正常乳腺组织之间相比差异均有显著性(P≤0.05,P<0.01)。在IDC中,三者表达均与腋窝淋巴结转移有关(P≤0.05,P<0.01),cyclinD1蛋白的表达与组织学分级有关(P<0.01),cyclinE蛋白的表达与肿块大小有关(P<0.01);p57kip2与cyclinD1之间、p57kip2与cyclinE之间的表达均呈负相关(P<0.01)、cyclinD1与cyclinE之间的表达呈正相关(P<0.01)。结论p57kip2蛋白低表达、cyclinD1和cyclinE蛋白高表达可能是乳腺组织恶性转变以及乳腺癌发生淋巴结转移的重要生物学标志,cyclinD1和cyclinE蛋白异常表达是乳腺癌发生的早期事件。联合检测p57kip2、cyclinD1及cyclinE蛋白对预测乳腺癌淋巴结转移有重要意义。
Purpose To investigate the effects of p57^(kip2),cyclin D1,and cyclin E on the development and progression of breast carcinomas. Methods Immunohistochemical S-P method was employed to detect expression of p57^(kip2),cyclin D1,and cyclin E proteins in 64 cases of IDC,15 cases of ductal carcinomas in situ (DCIS),and 15 cases of adjacent normal breast tissues. Results The expression of p57^(kip2),cyclin D1,and cyclin E showed significant difference(P≤0.05)between IDC and breast different tissues. Expression of cyclin D1,cyclin E also showed significant difference between DCIS and normal tissues (P<0.01). In IDC,there was correlation between the respective expression of three proteins with axillary lymph node metastasis(P≤0.05,P<0.01),between expression of cyclin D1 with histological grade(P<0.01),and between expression of cyclin E with tumor size(P<0.01). There was negative correlation between expression of p57^(kip2) and cyclin D1,and between expression of p57^(kip2)and cyclin E(P<0.01),but positive correlation between expression of cyclin D1 and cyclin E(P<0.01). Conclusions The decreased expression of p57^(kip2) and over-expression of cyclin D1,and cyclin E might be important biological markers for malignant transformation in breast tissue and lymph node metastasis of breast carcinomas,and the abnormal expression of cyclin D1,and cyclin E is early event of breast carcinomas. The combined detection of p57^(kip2)、cyclin D1,and cyclin E has positive effects on predicting the lymph node metastasis in breast carcinomas.
