摘要
目的 :改进一种测定血清总脂肪酸的气相色谱方法。方法 :用C17:0 做内标 ,用甲醇 /苯 (体积比 4/1)混合试剂及氯化乙酰对血清中总脂肪酸进行甲酯化反应 ,在DB -17石英毛细管柱上分离 ,用氢火焰离子检测器检测 ,分别用保留时间和峰面积定性、定量。通过摸索 ,在 70℃下衍生化 2h ,即能得到满意效果。结果 :本实验所用的 5种脂肪酸的色谱分离在 10min内就可完成 ,且各脂肪酸标准曲线的回归系数都在 0 99以上 ,不同浓度标准品的回收率也约在 97%~10 1%。结论 :该方法稳定可靠 ,易于操作重复 ,精确度及分辨率高。
Objective:To improve a gas chromatographic (GC) method for the determination of total fatty acids compositions in human serum Methods: Directly esterified by methanol/benzene (v/v:4/1) solution and acetyl chloride, the human total fatty acids were separated by a DB-17 quartz capillary column, using heptadecanoic acid (C 17∶0 ) as the internal standard (IS), detected by hydrogen flame ion-generation detector, and determined the nature and quantities by retention time and peak area ratio The methylation reaction was accomplished at 70℃ for 2 hours Results: The detection and separation of peaks for 5 tested fatty acids were performed within 10 min The regression coefficients of the standard curves of all the fatty acids were above 0 99 The recoveries of different fatty acids were about between 97%~101% Conclusion: The method is stable, reliable and simple, with a high precision and resolution [
