两种测定血浆非酯化脂肪酸方法的比较 被引量:5

A comparison of two different extraction methods for noncsterified fatty acids in serum and its clinical significance

摘要 亚油酸(C18:2)、亚麻油酸(C18:3)、花生四烯酸(C20:4)等不饱和脂肪酸对人体有重要的生物学作用。为了解输入脂肪乳剂后血中非酯化脂肪酸的变化,寻找比较简便、可靠的测定方法,选择近年文献上较常用的两种方法,层析加萃取法(Dole的方法),和直接萃取法(MacGee的方法)进行了对比研究。结果表明:两种方法的重复性误差用CV值(%)表示:Dole法为1.92~3.08;MacGee法为0.50~2.86。仪器的重复性误差CV(%)值是0.35~3.54。两种方法的加标回收率(%):Dole法是90.5~99.8;MacGee法是92.8~104。同一血样两种方法的测得值有可比性。Dole的方法提取时间较长、损失多,因此较难测到亚麻酸和花生四烯酸,但适用于分析细胞膜和组织中各成分的脂肪酸MacGee的方法取血少,步骤简单,能测定全部的必需脂肪酸。适用于测定血浆非酯化脂肪酸,可为早期必需脂肪酸缺乏提供依据。 To understand the changes of nonesterified fatty acid(NEFA) in serum after infusing lipid emulsion, two methods were compared. The methods compared were extraction plus thin layer chromatography(Dold method) and direct extraction (MacGee method)by capillary gas-liquid chro-matographic analysis. The reproducibility of two methods(CV) were 1. 92-3. 08(Dole) and 0. 5-2. 86(MacGee) respectively. Using Dole method linolenic acid and arachidonic acid are very difficult to be determine because of being time consmuing and more loss. But it is appropriate for determining fatty acids in the membrane of erythrocyte and tissue. MacGee mrthod is more simple and convenient and could measure all the fatty acid, it is suitable for determining NEFA in serum and might be useful in the diagnosis of early essential fatty acid deficiency.
