摘要
目的 探讨Aβ1-42 诱导模拟人类Alzheimer’s病 (AD)的大鼠病理模型中星形胶质细胞形态学变化及其在AD中的作用机制。方法 双侧杏仁核内注射 β-淀粉样多肽 1~ 4 2片段 (Aβ1-42 )制作大鼠AD模型 ,注射一周后采用GFAP免疫组化染色分析大鼠海马GFAP的表达。结果 杏仁核内注射Aβ1-42 后海马区出现星形胶质细胞增生、肥大、数目明显多于对照组 (P <0 .0 5 )。结论 AD模型神经元损伤、死亡与Aβ神经毒性直接相关 ,反应性星形胶质细胞参与Aβ导致神经元细胞毒性损伤作用。
Objective:To study the changes of astrocyte in the rat brain after injection of Aβ 1-42 into the amygdala. Methods:Microinjection of Aβ 1-42 into amygdala induced the rat learning and memory dysfunction. GFAP immunohistochemistry staining were employed. Results:The astroglia hypertrophy was seen in the rats injected with Aβ 1-42 in hippocampus, and the number of GFAP positive cells obviously increased compared with the controls (P<0.05).ConclusionAβ 1-42 induce neurons damage was direct correlated with the learning and memory deficits in the Alzheimer's disease model rats, reactive astrocyte participate in this cytotoxicity procedur, indirect cause the learning and memory deficits.
