摘要
目的 探讨miR-146a rs2910164位点单核苷酸多态性(single nucleotide polymerphism,SNP)与类风湿关节炎(rheumatoid arthritis,RA)病情活动及临床表现的相关性.方法 以高分辨率熔解(high-resolution melting,HRM)法对126例RA患者和102名正常对照进行miR-146a rs2910164位点的SNP分型,比较各基因型及等位基因频率在正常对照和RA患者中的差异,并分析其与RA患者病情活动性及临床表现之间的关系.结果 RA患者的miR-146a rs2910164位点G等位基因频率(30.6%)低于正常对照(32.8%),而C等位基因频率(69.4%)高于正常对照(67.2%);RA患者中GG基因型的比例(12.7%)低于正常对照(14.7%),而CC基因型的比例(51.6%)高于正常对照(49.O%),但差异均无统计学意义.具有GC基因型的RA患者出现关节外表现的比例显著增高(P<0.01),但是并未发现miR-146a rs2910164位点的不同基因型之间的DAS28评分的差异有统计学意义(P>0.01).结论 miR-146a rs2910164位点SNP非RA发病的独立风险因素,并且与RA病情活动性无明显相关性,但miR-146a rs2910164位点的GG基因型与RA特定的临床表现有关,提示miR-146a可能在RA发病中具有一定的作用.
Objective To assess the association of single nucleotide polymorphisms(SNPs)of microRNA-146a(miR-146a)with clinical features of rheumatoid arthritis(RA).Methods In 126 patients with RA and 102 matched healthy controls,SNPs of miR-146a rs2910164 locus were determined with a high-resolution melting method.The association of such polymorphisms with disease activity score in 28 joints(DAS28)and clinical features of RA was assessed.Results The distribution of SNPs of miR-146a rs2910164 among RA patients did not differ from that of the control group.No significant association was found between miR-146a rs2910164 polymorphism with DAS28.However,RA patients with a GG genotype had a higher chance to develop extraarticular manifestations(P<0.01).Conclusion Polymorphisms of miR-146a rs2910164 locus is not an independent risk factor for RA,though its GG genotype may be associated with extraarticular manifestations of RA.
