摘要
目的 探讨亚甲蓝光化学法病毒灭活和(或)滤除白细胞对冷沉淀凝血因子质量的影响。方法 选取陕西省血液中心2020年2月采集的400 ml全血60袋,随机分成冷沉淀组(对照组)、去白冷沉淀组、灭活冷沉淀组和去白灭活冷沉淀组,每组15袋,采用单因素方差分析比较4组凝血因子Ⅷ和FIB含量。结果 对照组、去白冷沉淀组和灭活冷沉淀组Ⅷ因子含量[(141.76±32.66)IU,(135.23±33.78)IU和(117.06±32.39)IU]均高于质量标准(≥80 IU),且符合率均>75.0%;去白灭活冷沉淀组凝血因子Ⅷ含量低于对照组[(101.09±37.47)IU比(141.76±32.66)IU],差异有统计学意义(P <0.05),符合率(73.3%)低于75.0%。4组FIB含量[(315.21±23.34)mg,(298.87±30.62)mg,(228.33±49.44)mg,(213.62±41.82)mg]均高于质量标准(≥150 mg),且符合率均>75.0%。结论 病毒灭活或滤除白细胞制备的凝血因子Ⅷ和FIB含量均符合国家相关标准,值得在临床推广应用。
Objective To explore the effects of virus inactivation by methylene blue photochemistry and/or filtration of leucocyte on cryoprecipitate coagulation factors.Methods A total of 60 bags of whole blood(400 ml per-bag)collected by Shannxi Blood Center were selected in February 2020,and were randomly divided into cryoprecipitate group(control group),leukocyte-reduced cryoprecipitate group,virus inactivated cryoprecipitate group,leukocyte-reduced and virus inactivated cryoprecipitate group,with 15 bags in each group.The contents of coagulation factor Ⅷ and FIB in the four groups were compared by one-way variance analysis.Results The contents of coagulation factor Ⅷ in the control group,leukocytereduced cryoprecipitate group,virus inactivated cryoprecipitate group[(141.76±32.66)IU,(135.23±33.78)IU,(117.06±32.39)IU]were all higher than the quality standard(≥80 IU),and the qualified rate was all higher than 75.0%.The contents of coagulation factor Ⅷ in the leukocyte-reduced and virus inactivated cryoprecipitate group was lower than that in the control group,with statistical significance[(101.09±37.47)IU vs.(141.76±32.66)IU,P<0.05],and the qualified rate(73.3%)was lower than 75.0%.The contents of FIB in the four groups[(315.21±23.34)mg,(298.87±30.62)mg,(228.33±49.44)mg,(213.62±41.82)mg]were all higher than the quality standard(≥150 mg),and the coincidence rates were all higher than 75%.Conclusions The contents of coagulation factor Ⅷ and FIB of cryoprecipitate are in line with the national standards after virus inactivated or leukocyte-reduced treatment,and are worth popularizing in clinical application.
