犀角地黄汤调控银屑病PI3K/Akt/mTOR致病通路的作用机制研究 被引量:4

Study on mechanism of Xijiao Dihuang Decoction regulating pathogenic pathway PI3K/Akt/mTOR in psoriasis

摘要 目的:探讨犀角地黄汤调控银屑病PI3K/Akt/mTOR致病通路的作用机制。方法:将60只BALB/c小鼠随机分为6组:正常组、模型组、阳性药物组、犀角地黄汤低、中、高剂量组。除正常组涂抹凡士林外,其余各组每日在小鼠裸露背部涂抹5%咪喹莫特(IMQ)乳膏,间隔4小时后,各组给予相应药物进行干预。采用银屑病皮损严重程度指数评分(PASI)评价小鼠背部皮损严重程度;HE染色检测小鼠皮损组织病理学变化;Western Blot法检小鼠银屑病样皮损PI3K/Akt/mTOR致病通路中PI3K(p^(85))、PI3K(p^(110α))、Akt、p-Akt(S^(473))、p-Ak(Thr^(308))、mTOR、p-mTOR(Ser^(2448))、p-mTOR(Ser^(2481))、P70S6K1、p-p70S6K(Thr^(389))等关键蛋白的表达情况。结果:与模型组相比,犀角地黄汤各剂量组和阳性药物雷帕霉素(RPM)组银屑病样皮损显著减轻,PASI评分降低,差异有统计学意义(P<0.05)。Western Blot结果显示,与模型组比较,犀角地黄汤各剂量组PI3K/Akt/mTOR通路相关蛋白表达水平显著降低,差异有统计学意义(P<0.05)。与雷帕霉素组比较,犀角地黄汤高、中剂量组PI3K/Akt/mTOR致病通路中相关蛋白的表达水平相近(P>0.05)。结论:犀角地黄汤可以降低小鼠银屑病样皮损中PI3K/Akt/mTOR致病通路的相关蛋白表达,这可能与其发挥治疗银屑病临床疗效有关。 Objective:To investigate the mechanism of Xijiao Dihuang Decoction regulating pathogenic pathway PI3K/Akt/mTOR in psoriasis.Method:A total of 60 BALB/c mice were randomly divided into 6 groups:normal group,model group,positive drug group and low-,medium-and high-dose groups of Xijiao Dihuang Decoction.The normal group was smeared with vaseline,and the other groups were smeared with 5%imiquimod(IMQ)cream on the bare back of mice every day.After 4 hours,each group was given corresponding drugs for intervention.The severity of back lesions of mice was evaluated by psoriasis area and severity index(PASI),the histopathological changes of mouse skin lesions were detected by HE staining,and Western Blot was conducted to test the key proteins of pathogenic pathway PI3K/Akt/mTOR in psoriatic lesions,including PI3K(p^(85)),PI3K(p^(110α)),Akt,p-Akt(S^(473)),p-Ak(Thr^(308)),mTOR,p-mTOR(Ser^(2448)),p-mTOR(Ser^(2481)),P70S6K1 and p-p70S6K(Thr^(389)).Result:Compared with the model group,the psoriatic lesions and PASI scores in each dose group of Xijiao Dihuang Decoction and the positive drug rapamycin(RPM)group were significantly reduced(P<0.05).The results of Western Blot showed that the levels of the related proteins of PI3K/Akt/mTOR pathway in each dose group of Xijiao Dihuang Decoction were significantly decreased compared with the model group,with significant differences(P<0.05).Compared with RPM group,the expression levels of related proteins of the PI3K/Akt/mTOR pathway in high-and medium-dose groups of Xijiao Dihuang Decoction were similar(P>0.05).Conclusion:Xijiao Dihuang Decoction can reduce the expression of related proteins of the pathogenic pathway PI3K/Akt/mTOR in psoriatic lesions of mice,which may be related to its clinical efficacy in the treatment of psoriasis.
