电针干预脊髓损伤大鼠督脉脊髓受损节段的神经营养因子NGF、TrkA的表达 被引量:5

摘要 背景:中医治疗脊髓损伤有“从督脉论治”的理论基础,但其作用机制以及对脊髓损伤后神经营养因子NGF、TrkA的调控尚且不详。目的:通过观察电针干预脊髓损伤(SCI)大鼠督脉脊髓受损节段的mRNA和神经营养因子NFG、TrkA的表达,研究电针督脉对脊髓损伤大鼠受损节段的干预机制。方法:将60只大鼠随机分为假手术组、损伤对照组、督脉组,采取脊髓半横断法进行造模,采用电针督脉的方式进行干预治疗,并分别在干预前3天,干预后7、14、21天(即第3、10、17、24天)肢功能评分(BBB评分)及取材。然后将得到脊髓组织进行实时荧光定量检测、Western blot检测、HE染色检测。结果与结论:(1)BBB评分结果:督脉组评分高于损伤对照组,差异均有显著性意义(P<0.05);(2)实时荧光定量PCR结果:NGF、TrkA基因在督脉组在各时间点均有不同程度的升高,差异显著,有统计学意义(P<0.05);(3)Western blot检测结果:NGF、TrkA基因在督脉组在各时间点均有不同程度的升高,差异显著,有统计学意义(P<0.05);(4)HE染色检测:督脉组10天出血损伤有所减轻,出现大量脂肪空泡及少量炎细胞浸润;随着时间推移,损伤逐渐减轻,但24天时仍有少量神经元轻微固缩。结论:电针针刺督脉能够改善脊髓损伤大鼠的BBB评分结果,上调NGF、TrkA因子的表达,进而促进神经细胞的修复。 Objective:To observe the mRNA and the expression of neurotrophic factors NFG and TrkA in the injured segment of spinal cord of Du meridian in rats with spinal cord injury(SCI),and to study the intervention mechanism of Du meridian on the injured segment of spinal cord injury in rats.Methods:60 rats were randomly divided into a sham operation group,a injury controlled group and a Du meridian group.The model was made by semi transection of spinal cord.The intervention treatment was performed by Electroacupuncture of Du meridian.The limb function score(BBB score)and materials were obtained 3 days before the intervention and 7,14 and 21 days after the intervention(i.e.days 3,10,17 and 24).Then the obtained spinal cord tissue was detected by real-time fluorescence quantitative detection,Western blot detection and HE staining.Results:(1)BBB score:the score of the Du meridian group was significantly higher than that of the injury controlled group(P<0.05).(2)Results of real-time fluorescence quantitative PCR:NGF and TrkA genes in the Du meridian group increased in varying degrees at each time point,with significant difference(P<0.05).(3)Results of Western blot:NGF and TrkA genes in the Du meridian group increased in varying degrees at each time point,with significant difference(P<0.05).(4)He staining showed that the bleeding injury was alleviated,and a large number of fat vacuoles and a small amount of inflammatory cell infiltration appeared in the Du meridian group at 10 days.With the passage of time,the injury decreased gradually,but a small number of neurons were slightly pyknotic at 24 days.Conclusion:The results show that electroacupuncture at Du meridian can improve the BBB score of spinal cord injury rats,up regulate the expression of NGF and TrkA factors,and then promote the repair of nerve cells.
