摘要
目的:基于网络药理学研究半夏泻心汤治疗慢性萎缩性胃炎的作用机制。方法:利用TCMSP数据库,得到半夏泻心汤成分的口服利用率和药物相似性等参数,并筛选药物的有效成分组,收集有效成分的靶点。通过Dis Ge NET数据库得到慢性萎缩性胃炎的疾病靶点,在与半夏泻心汤靶点取得交集后得到了药效靶点。将上述靶点输入到String数据库进行蛋白质-蛋白质相互作用网络预测,最后利用DAVID数据库进行KEGG富集分析,阐明半夏泻心汤治疗慢性萎缩性胃炎的潜在信号通路。结果:我们筛选出了半夏泻心汤194个活性成分和154个作用靶点。通过蛋白质互作网络分析,半夏泻心汤治疗慢性萎缩性胃炎可能与BCL-2,PTGS2,TNF,GSTP1,GSTM1等靶点有关。KEGG富集分析结果显示半夏泻心汤治疗慢性萎缩性胃炎与TNF信号通路,NF-κB信号通路,NOD样受体信号通路,PI3k/Akt信号通路,Toll样受体信号通路等有关。结论:半夏泻心汤治疗慢性萎缩性胃炎具有中药复方多成分-多靶点-多通路的特点,其治疗慢性萎缩性胃炎的生物学机制可能与TNF signaling pathway等信号通路有关。
Objective:To study the mechanism of Banxia Xiexin Decoction in the treatment of chronic atrophic gastritis based on network pharmacology.Methods:Using the TCMSP database,the oral utilization rate and drug similarity of the ingredients of Banxia Xiexin Decoction were obtained,and the effective component groups of the drug pairs were screened to collect the target of the active ingredients.The disease target of chronic atrophic gastritis was obtained through the Dis Ge NET database,and the drug effect target was obtained after the intersection with the target of Banxia Xiexin Decoction.The above target was input into the String database for protein-protein interaction network prediction.Finally,the DAVID database was used for KEGG enrichment analysis to clarify the potential signaling pathway of Banxia Xiexin Decoction in the treatment of chronic atrophic gastritis.Results:We screened 194 active ingredients and 154 targets for Banxia Xiexin Decoction.Through protein interaction network analysis,Banxia Xiexin Decoction for chronic atrophic gastritis may be related to BCL-2,PTGS2,TNF,GSTP1,GSTM1 and other targets.The results of KEGG enrichment analysis showed that Banxia Xiexin Decoction was involved in the treatment of chronic atrophic gastritis with TNF signaling pathway,NF-κB signaling pathway,NOD-like receptor signaling pathway,PI3 k/Akt signaling pathway,Toll-like receptor signaling pathway.Conclusion:Banxia Xiexin Decoction has the characteristics of multi-component-multi-target-multi-channel of traditional Chinese medicine compound in treating chronic atrophic gastritis.The biological mechanism of treatment of chronic atrophic gastritis may be related to signaling pathways such as TNF signaling pathway.
