基于“脑肠互动”探讨和胃理气方治疗肝郁型功能性消化不良机制研究 被引量:10

Study the Mechanism of Model Rats of Functional Dyspepsia with Liver Depression Treated by Hewei Liqi Decoction(和胃理气方)Based on Brain-gut Interaction

摘要 目的观察和胃理气方对肝郁型功能性消化不良模型大鼠脑组织中脑源性神经营养因子(BDNF)、环磷腺苷效应元件结合蛋白(CREB)的影响,从分子水平探讨其作用机制。方法选取40只健康Wistar SPF级大鼠,经过随机数字表法将其随机分为空白组、模型组、中药组和西药组4组,每组10只。采用改良式CUMS方法建立肝郁型功能性消化不良模型;分别用和胃理气方和吗丁啉进行干预治疗。通过敞箱、糖水偏好实验评价大鼠抑郁状态;采用胃排空、小肠推进实验评价胃肠运动情况;采用Western blot法测定大鼠海马体中CREB及p-CREB蛋白表达;采用ELISA法测定大鼠海马体中BDNF含量。结果(1)与空白组比较,模型组大鼠糖水消耗率下降,敞箱实验水平及垂直得分下降;胃排空及小肠推进减慢;海马体中CREB、p-CREB蛋白表达下降,BDNF含量下降,有统计学意义(P<0.05)。(2)治疗后与模型组比较,中药组和西药组糖水消耗率提高,敞箱实验水平及垂直得分增加;海马体中CREB、p-CREB蛋白表达升高,BDNF含量升高,有统计学意义(P<0.05)。(3)治疗后,中药组和西药组海马体中CREB、p-CREB蛋白表达无统计学意义(P>0.05);中药组海马体中BDNF含量高于西药组,有统计学意义(P<0.05)。结论肝郁可致"脑肠轴"失衡,和胃理气方可以改善FD肝郁大鼠抑郁状态,促进胃肠动力,其分子机制与上调CREB/BDNF信号通路相关蛋白表达有关。 Objective To observe the effect of brain-derived neurotrophic factor(BDNF)and cyclicamp response element binding protein(CREB)of model rats suffered from functional dyspepsia with liver depression by Hewei Liqi Decoction(和胃理气方),probe into its mechanism from molecular level.Methods Selected 40 healthy Wistar SPF rats,divided the rats with random number table method into blank group,model group,Chinese herb group,Western drug group,and 10 each group.Adopted the improved method of chronic unpredictable mild stress(CUMS)to set the model rats of functional dyspepsia with liver depression,intervention treatment with Hewei Liqi Decoction and Domperidone respectively.Evaluated whether the rat was in depressive state by open-field test and the consumption of saccharose;Assessed the state of gastrointestinal motility by the experimental methods of gastric emptying and small intestine propulsion.Measured the expression quantity of CREB,p-CREB protein from hippocampus by Western-blot testing;Measured the expression quantity of BDNF from hippocampus by ELISA testing.Results(1)Compared with blank group,in model group,saccharose consumption rate went down,openfield testing level and vertical score went down;gastric emptying and small intestine propulsion slowed down;the expression quantity of CREB,p-CREB protein from hippocampus went down;the expression quantity of BDNF from hippocampus went down;with statistical difference(P<0.05).(2)Compared with model group after treatment,in Chinese herb group and Western drug group,saccharose consumption rate went up,open-field testing level and vertical score went up;the expression quantity of CREB,p-CREB protein from hippocampus went up;the expression quantity of BDNF from hippocampus went up,with statistical difference(P<0.05).(3)After treatment,compared Chinese herb group with Western drug group,there was no statistical difference in the result of expression quantity of CREB,p-CREB protein from hippocampus(P>0.05);there was statistical difference in the result of expression quantity of BDNF from hippocampus(P<0.05).Conclusion Liver depression can lead to imbalance of the brain-gut-axis,Hewei Liqi Decoction can improve depression state of FD rats with liver depression,promote gastrointestinal motility,its molecular mechanism is related to the expression of relevant protein of up-regulating CREB/BDNF signal path.
