生、制白术制枳术丸对功能性消化不良大鼠“脑-肠”轴功能影响的研究 被引量:14

Study of Zhizhu Pill(枳术丸)Preparation by Raw and Processed Baizhu(Atractylodes macrocephala)on Functional Dyspepsia Rats of Brain-Gut Axis

摘要 目的从对“脑-肠”轴功能影响的角度探讨枳术丸治疗功能性消化不良(FD)的机制,并比较生制白术制枳术丸的作用差异。方法采用夹尾刺激联合不规则饮食复合因素建立FD大鼠模型,阳性药组给予成人等效剂量的莫沙必利(6.25 mg·kg^(-1))及益生菌(0.83 g·kg^(-1)),中药干预组给予不同剂量的生制白术制枳术丸(2.625、5.25、10.5 g·kg^(-1))。给药2周,观察各组大鼠的一般状态;实验结束,测定各组大鼠的体质量、胃排空率、肠推动率,并以酶联免疫吸附法(ELISA)检测各组大鼠血清中神经递质5-羟色胺(5-HT)、皮质醇(CORT)、血管活性肠肽(VIP)及胃肠激素降钙素基因相关肽(CGRP)、胆囊收缩素(CCK)、酪酪肽(PYY)的水平;以硝酸还原酶法测定大鼠血清中神经递质一氧化氮(NO)的水平。结果与模型组相比,生、制白术制枳术丸均可显著提高FD大鼠的体质量、胃排空率、肠推动率,调整模型动物血清中神经递质及胃肠激素的水平(P<0.05),并且制白术制枳术丸的作用稍强于生白术制枳术丸。结论枳术丸可通过对“脑-肠”轴功能的调节来治疗功能性消化不良,并且使用制白术进行组方配伍效果更佳。 Objective To research the mechanism of pharmacodynamic effect of Zhizhu Pill(枳术丸)in functional dyspepsia rats from the“brain-gut”axis and compare the difference in effect of Zhizhu Pill preparation by raw and processed Baizhu(Atractylodes macrocephala)on functional dyspepsia rats.Method The model of functional dyspepsia rat was used complex factors of clip-tail stimulation combine with irregular dietary.The model animal was given the clinical equivalent dose of positive drug mosapride(6.25 mg·kg^(-1))or probiotics(0.83 g·kg^(-1))and different dosage Zhizhu Pill preparation by raw and processed(2.625,5.25,10.5 g·kg^(-1)).The general state of rats in each group was observing after model 2 weeks.Then,it was measured in the rates of gastric emptying and intestinal propulsion in each group after the experiment.The levels of neurotransmitter which included 5-HT,CORT,VIP and gastrointestinal hormones which included CGRP,CCK and PYY,in the serum were measured by ELISA.The level of NO which was a neurotransmitter in the serum was measured by nitrate reductase method.Results Compared to those of the model group,both Zhizhu Pill preparation by raw and processed Baizhu(Atractylodes macrocephala)could improve the body mass,the rates of gastric emptying and intestinal propulsion,the levels of neurotransmitter and gastrointestinal hormones on functional dyspepsia rats(P<0.05).However,it is superior that the regulating effect of Zhizhu Pill preparation by the processed Baizhu(Atractylodes macrocephala)in the body mass,the rates of gastric emptying and intestinal propulsion,the level of neurotransmitter and gastrointestinal hormones on functional dyspepsia rats.Conclusion Zhizhu Pill could influence the"brain-gut"axis to treat functional dyspepsia rats and it is more reasonable to use processed Baizhu(Atractylodes macrocephala)in Zhizhu Pill.
