摘要
外泌体是一类源于细胞内膜,大小在30~100 nm的囊泡。囊泡内包含多种蛋白质和核酸等。其在细胞之间的信息通讯中具有关键的作用。在包含的多种RNA中,MicroRNAs(miRNAs)是一类高度保守的,长度由19~25个核苷酸组成的微小内源性非编码RNA。它可通过靶向mRNA的3’非翻译区(3’UTR)来介导靶基因转录后的基因沉默,从而起到调节基因表达的作用。近年来的研究表明,miRNAs参与了动脉粥样硬化的形成。它们通过多靶点、多途径的特点影响体内脂肪代谢、炎症反应以及细胞的增殖、分化、凋亡和自噬,从而促进或抑制疾病的发生发展。中医学凭借多靶点、多环节的调节特点,在动脉粥样硬化的临床治疗中具有可观的疗效。中医药基于调控miRNAs表达量对动脉硬化的防治受到广泛关注。文章就miRNAs在动脉粥样硬化形成过程的机制,以及中医学干预动脉硬化的发生发展进行综述。
Exosomes are a type of vesicles that originate from the inner cell membrane and are between 30 and 100 nm in size.Vesicles contain a variety of proteins and nucleic acids.It plays a key role in information communication between cells.Among the various RNAs included,microRNAs(miRNAs)are a class of highly conserved,tiny endogenous non-coding RNAs composed of 19-25 nucleotides in length.It can mediate the post-transcriptional gene silencing of target genes by targeting the 3’untranslated region(3’UTR)of mRNA to regulate gene expression.Recent studies have shown that miRNAs are involved in the formation of atherosclerosis.They affect fat metabolism,inflammatory response and cell proliferation,differentiation,apoptosis and autophagy through the characteristics of multiple targets and multiple pathways,thereby promoting or inhibiting the occurrence and development of diseases.Relying on the multi-target and multi-step regulation characteristics,the Chinese medicine has considerable curative effect in the clinical treatment of atherosclerosis.The prevention and treatment of diseases based on the regulation of miRNA expression in traditional Chinese medicine has received widespread attention.This article reviews the mechanism of miRNAs in the process of atherosclerosis and the development of the intervention of traditional Chinese medicine.
