LncRNA SNHG20和miR-495在胆囊癌中的表达及临床意义 被引量:2

Expression and clinical significance of LncRNA SNHG20 and miR-495 in gallbladder carcinoma

摘要 目的研究胆囊癌中长链非编码RNA(LncRNA)SNHG20和miR-495表达变化及其相关性,并分析LncRNA SNHG20和miR-495表达水平与患者预后的关系。方法选择2015-2018年手术切除的121例胆囊癌患者作为研究对象,术中同时留取患者肿瘤组织和距肿瘤5 cm外的癌旁组织,采用RT-PCR法分别检测肿瘤组织和癌旁组织中SNHG20和miR-495表达水平,分析肿瘤组织中SNHG20和miR-495表达水平的相关性及其与患者临床病理特征和预后的关系。结果肿瘤组织SNHG20相对表达量显著高于癌旁组织,miR-495相对表达量显著低于癌旁组织,差异均有统计学意义(P<0.05);肿瘤组织中SNHG20和miR-495表达水平呈显著负相关关系(r=-0.517,P=0.001);肿瘤组织中SNHG20相对表达量与患者淋巴结转移、肿瘤大小、局部浸润深度有关,miR-495相对表达量与患者淋巴结转移、肿瘤分化程度、TNM分期、局部浸润深度有关;118例患者得到有效随访,平均随访时间(22.37±6.52)个月,其中死亡83例,存活35例,3年总生存率29.66%。死亡组患者肿瘤组织中SNHG20相对表达量显著高于生存组,miR-495相对表达量显著低于生存组,差异均有统计学意义(P<0.05);SNHG20表达量≥2.06组患者3年生存率显著低于SNHG20<2.06组患者,差异均有统计学意义(P<0.05);miR-495≥0.53组患者3年生存率显著高于miR-495<0.53组患者,差异均有统计学意义(P<0.05);淋巴结转移、TNM分期、肿瘤分化程度、SNHG20高表达及miR-495低表达是影响胆囊癌预后的独立危险因素。结论LncRNA SNHG20与miR-495在胆囊癌组织中表达呈负相关,SNHG20可能通过调控miR-495参与胆囊癌发生发展过程,是影响患者预后不良的危险因素。 Objective To study the changes and correlation of long non-coding RNA(LncRNA)SNHG20 and miR-495 expression in gallbladder carcinoma,and analyze the relationship between the expression level of LncRNA SNHG20 and miR-495 and the prognosis of patients.Methods 121 cases of gallbladder carcinoma from 2015 to 2018 were selected as the subjects.The tumor tissues and normal tissues adjacent to the tumor 5 cm from the tumor were retained during the operation.The expression of SNHG20 and miR-495 in the tumor tissue and the normal tissues adjacent to the cancer was detected by RT-PCR method,and the correlation between SNHG20 and miR-495 expression in tumor tissues and their relationship with clinicopathological features and prognosis were analyzed.Results The relative expression of SNHG20 in tumor tissues was significantly higher than that in adjacent normal tissues(P<0.05),and the relative expression of miR-495 was significantly lower than that in adjacent normal tissues(P<0.05);there was a significant negative correlation between the expression level of SNHG20 and miR-495 in tumor tissues(r=-0.517,P=0.001);the relative expression of SNHG20 in tumor tissues was related to lymph node metastasis,tumor size and local infiltration depth,and the relative expression of miR-495 was related to lymph node metastasis,tumor differentiation,TNM staging,and local infiltration depth.118 patients were followed up effectively,with an average follow-up time of(22.37±6.52)months,of which 83 cases died,35 cases survived,and 3 years overall survival rate was 29.66%.The relative expression of SNHG20 in the tumor tissues of the death group was significantly higher than that of the survival group(P<0.05),and the relative expression of miR-495 was significantly lower than that of the survival group(P<0.05).The 3 year survival rate of patients with SNHG20 expression exceeding 2.06 was significantly lower than those of patients with SNHG20<2.06(P<0.05).The 3 year survival rate in group miR-495≥0.53 was significantly higher than that in group miR-495<0.53(P<0.05).Lymph node metastasis,TNM stage,tumor differentiation,high expression of SNHG20 and low expression of miR-495 were independent risk factors for the prognosis of gallbladder carcinoma.Conclusion The expression of LncRNA SNHG20 and miR-495 in the carcinoma of gallbladder were negatively correlated,and SNHG20 might be involved in the development of gallbladder cancer by regulating miR-495,and it was a risk factor that could affect the poor prognosis of the patients.
