摘要
【目的】探讨长链非编码RNA(LncRNA)Fez家族锌指蛋白1反义RNA1(lncRNA FEZF1-AS1)在胆囊癌患者中的表达水平及其与预后关系。【方法】回顾性选取本院2015年11月至2017年3月诊治的66例胆囊癌患者的癌组织及癌旁胆囊黏膜组织进行研究。采用实时荧光定量PCR(qRT-PCR)法检测胆囊癌组织及癌旁胆囊黏膜组织LncRNA FEZF1-AS1表达水平;分析癌组织LncRNA FEZF1-AS1表达水平与胆囊癌患者临床病理特征的关系;采用Kaplan-Meier曲线法分析胆囊癌患者癌组织中LncRNA FEZF1-AS1表达水平与胆囊癌患者预后的关系;采用COX回归分析胆囊癌预后的影响因素。【结果】胆囊癌组织LncRNA FEZF1-AS1表达水平明显高于癌旁正常组织(P<0.05);胆囊癌患者癌组织LncRNA FEZF1-AS1表达水平与胆囊癌患者TNM分期、淋巴结转移、分化程度、浸润深度相关(P<0.05),而与患者性别、年龄、肿瘤大小、位置无关(P>0.05);胆囊癌患者LncRNA FEZF1-AS1高表达组术后36个月生存率明显低于LncRNA FEZF1-AS1低表达组(P<0.05);TNM分期、淋巴结转移、LncRNA FEZF1-AS1是影响胆囊癌患者预后的独立危险因素(P<0.05)。【结论】LncRNA FEZF1-AS1在胆囊癌患者癌组织中表达水平明显升高,且高水平LncRNA FEZF1-AS1与胆囊癌患者不良预后密切相关,可用于判定胆囊癌患者预后情况。
【Objective】To investigate the expression level of long non-coding RNA(LncRNA)Fez family zinc finger 1-antisense RNA1(LncRNA FEZF1-AS1)in patients with gallbladder cancer and its relationship with the prognosis.【Methods】The cancer tissues and adjacent gallbladder mucosal tissues in 66 patients with gallbladder cancer treated in our hospital from November 2015 to March 2017 were retrospectively studied.Real-time fluorescent quantitative PCR(qRT-PCR)was used to detect the expression level of LncRNA FEZF1-AS1 in gallbladder cancer tissues and adjacent gallbladder mucosal tissues.The relationship between the expression level of LncRNA FEZF1-AS1 and clinicopathological characteristics of gallbladder cancer was analyzed.Kaplan-Meier curve was used to analyze the relationship between LncRNA FEZF1-AS1 expression level and prognosis of gallbladder cancer.In addition,COX regression was used to analyze the prognostic factors of gallbladder cancer.【Results】The expression level of LncRNA FEZF1-AS1 in gallbladder cancer tissues was significantly higher than that in paracancerous normal tissues(P<0.05).The expression level of LncRNA FEZF1-AS1 in gallbladder cancer was correlated with TNM stage,lymph node metastasis,differentiation degree and infiltration depth(P<0.05),but not with gender,age,tumor size and location(P>0.05).The 36-month survival rate of gallbladder cancer in high expression group of LncRNA FEZF1-AS1 was significantly lower than that in low expression group of LncRNA FEZF1-AS1(P<0.05).TNM stage,lymph node metastasis and LncRNA FEZF1-AS1 were independent risk factors for the prognosis of gallbladder cancer(P<0.05).【Conclusion】The expression level of LncRNA FEZF1-AS1 in the cancer tissues of patients with gallbladder cancer is significantly increased,and the high level of LncRNA FEZF1-AS1 is closely related to the poor prognosis of patients with gallbladder cancer,which can be used to determine the prognosis of patients with gallbladder cancer.
