Ki-67和TOPOⅡα在女性乳腺浸润性导管癌中的表达及临床意义 被引量:2

Expression of Ki-67,TOPOⅡα in breast invasive ductal carcinoma and its clinical significance

摘要 目的探讨细胞核相关抗原Ki-67、DNA拓扑异构酶Ⅱα(TOPOⅡα)在女性乳腺浸润性导管癌组织中的表达及临床意义。方法收集2019年2月-2021年11月南京医科大学第四附属医院女性乳腺浸润性导管癌组织标本191例,采用免疫组织化学法检测Ki-67、TOPOⅡα在乳腺癌组织中的表达情况。乳腺癌分子分型采用免疫组化及荧光原位杂交(FISH)检测确定。结果女性乳腺浸润性导管癌组织中Ki-67的阳性表达率为64.9%(124/191),与浸润癌最大径、组织学分级、脉管侵犯及分子分型相关,差异均有统计学意义(χ^(2)=11.085、33.203、8.515、124.765,P均<0.05)。TOPOⅡα的阳性表达率为76.4%(146/191),与浸润癌最大径、组织学分级、临床分期、脉管侵犯及分子分型相关,差异均有统计学意义(χ^(2)=18.324、29.365、6.915、11.724、53.645,P均<0.05)。TopoⅡα与Ki-67共同表达与浸润癌最大径、组织学分级、脉管侵犯及分子分型相关,差异均有统计学意义(χ^(2)=17.545、43.561、13.690、104.416,P均<0.05)。Spearman相关分析显示,Ki67与TOPOⅡα在女性乳腺浸润性导管癌组织中表达成正相关(r=0.471,P<0.001)。结论Ki-67和TOPOⅡα与乳腺癌的发展相关,可以作为其判断预后的参考指标。 Objective To investigate the expression and clinical significance of nuclear-associatedantigen Ki-67,topoisomeraseⅡα(TOPOⅡα)in invasive female ductal breast carcinoma.Methods A total of 191 specimens of breast invasive ductal carcinoma were collected in the Fourth Affiliated Hospital of Nanjin Medical University from February 2019 to November 2021,and the expression of Ki-67,TOPOⅡαin breast cancer was detected by immunohistochemistry.Molecular typing of breast cancer was determined by immunohistochemistry and fluorescence in situ hybridization(FISH)detection.Results The positive expression rate of Ki-67 in breast invasive ductal carcinoma was 64.9%(124/191),which was associated with the maximum diameter of invasive carcinoma,histological grade,lympholymphovascular invasion and molecular type,and the differences were statistically significant(χ^(2)=11.085,33.203,8.515,124.765;P all<0.05).The positive expression rate of TOPOⅡαin breast invasive ductal carcinoma was 76.4%(146/191),which was associated with the maximum diameter of invasive carcinoma,histological grade,clinical stage,lymphovascular invasion and molecular subtype,and the differences were statistically significant(χ^(2)=18.324,29.365,6.915,11.724,53.645;P all<0.05).Co-expression of TOPOⅡαand Ki-67 was associated with maximum diameter of invasive carcinoma,histological grade,lymphovascular invasion and molecular subtype(χ^(2)=17.545,43.561,13.690,104.416;P all<0.05).Spearman correlation analysis showed that the expression of Ki67 and TOPOⅡαin the female breast invasive ductal carcinoma group was positively correlated(r=0.471,P<0.001).Conclusion Ki-67 and TOPOⅡαwere related to the development of breast cancer and which could be used as a reference index to judge its prognosis.
