摘要
目的借助中药网络药理学方法,探究五子衍宗丸在治疗不育、早泄、阳痿这3种疾病过程中的共性作用机制。方法五子衍宗丸相关活性成分的化学结构和药物靶点是利用中药系统药理学数据库与分析平台(TCMSP)得到的;上述3种疾病的相关靶点是利用人类孟德尔遗传数据库(OMIM),人类基因数据库(GeneCards)筛选得到的;利用Cytoscape 3.7.1构建"化合物-靶点-疾病"网络;再利用蛋白质相互作用(PPI)网络筛选出核心靶点,而基因本体(GO)功能富集分析和基于京都基因与基因组百科全书(KEGG)通路富集分析是借助生物学信息注释数据库(DAVID)进行。结果分析所构建的"疾病-基因-药物"网络,发现甾体类化合物是五子衍宗丸治疗不育、早泄、阳痿三种疾病的主要化学成分,再分析蛋白相互作用网络,得出五子衍宗丸治疗上述三种疾病的10个高频共性靶点,30个GO高频共性基因,6条KEGG高频共性通路。结论利用网络药理学方法探究五子衍宗丸在不育、早泄、阳痿这3种疾病治疗过程中的共性作用机制以及高频共性靶点、GO、KEGG通路等,为其活性成分研究和临床应用提供理论依据。
Objective To explore the common mechanism of Wuzi Yanzong Pill in the treatment of Sterility,Erectile Dysfunction and Premature Ejaculation,by network pharmacology method.Methods Wuzi Yanzong Pill active ingredients’relevant chemical structure and corresponding drug targets were obtained by the Traditional Chinese Medicine Systems Pharmacology Database(TCMSP);Three diseases related targets were screened by Online Mendelian Inheritance in Man(OMIM)and The Human Gene Database(GeneCards);The"herb-target-gene"networks were constructed by Cytoscape 3.7.1;The core targets were screened by Protein-Protein Interaction Networks(PPI);Gene Ontology(GO)function enrichment and Kyoto Encyclopedia of Genes and Genomes(KEGG)enrichment were analyzed by the Database for Annotation,Visualization and Integrated Discovery(DAVID).Results We concluded that the Steroid compounds were the main chemical component by"herb-gene-drug"networks,and 10 high frequency common targets,30 GO high frequency common genes and 6 KEGG high frequency common pathways were obtained at the same time.Conclusion In terms of the common mechanism,common high-frequency targets,genes,KEGG pathway between Wuzi Yanzong Pill and three diseases,Sterility,Erectile Dysfunction and Premature Ejaculation provided by network pharmacology method,our research provided theoretical basis for Wuzi Yanzong Pill’s active ingredient research and clinical application.
