基于性状与化学成分变化规律分析枯芩最佳采收年限 被引量:3

摘要 目的通过分析生长年限与黄芩性状和化学成分变化的关联规律,为枯芩合理采收年限的确定提供实验依据。方法在同一道地产区采收2年生子芩和3、5、7、10年枯芩,比较不同年限枯芩的性状差异。运用Matlab软件计算饮片中空腐朽面积占比,采用色差仪测定药材粉末色差值,采用超高液相色谱法考察成分整体特征及主要化学成分含量。结果随着采收年限的增加,枯芩中空腐朽面积逐渐增大;药材粉末颜色绿红值a^(∗)升高,亮度值L^(∗)、黄蓝值b^(∗)和总色差值ΔE^(∗)降低;生长年限差距越大,UPLC图谱的相似度降低越明显,由0.999降至0.907;黄芩苷和汉黄芩苷等黄酮苷类成分的含量逐年递减,黄芩素、汉黄芩素和千层纸素A等黄酮苷元类成分的含量逐年递增,5种成分总含量逐年递减。7年以上样品的色差值和化学成分变化趋于稳定。Pearson相关性分析表明,采收年限与黄芩腐朽面积、颜色值、化学成分含量显著相关。结论基于采收年限与化学成分含量、成分转变、腐朽面积、颜色变化规律综合评判,认为枯芩在3年以上7年以内采收较合理。 Objective By analyzing the correlation law between growth years and the changes of Scutellaria baicalensis characteristic and the chemical components,We provide an experimental basis for determining the reasonable harvesting years of Scutellaria baicalensis.Methods 2-year-old Ziqin and 3-,5-,7-,and 10-year-old Kuqin were harvested from the same production area,to compare difference of characteristic in different years of Kuqin.Matlab software was used to calculate the proportion of hollow decay area,and a colorimeter was used to measure the color difference as the two medicinal materials were shaped into powder.Ziqin and Kuqin with different ages were investigated overall characteristics of components and contents of main chemical components by ultra-high liquid chromatography.Results The results showed that with the increase of harvesting years,the di-ameter of all batch of decoction pieces continue to increase;the green-red numercial value a^(∗)of the two medicinal material’s powder color increases,the numercial brightness value L^(∗),the yellow-blue value b^(∗)and the total color difference numercial value ΔE^(∗)decreased.The greater the difference in growth years,the more significant the decrease in similarity of UPLC profiles,from 0.999 to 0.907.The contents of flavonoid glycosides such as baicalin and Wogonoside decrease year by year,the contents of baicalein,wogonin,oroxylin A increased,and eventually the total content of the five chemical components decreased,the color difference values and chemical composition changes of samples older than 7 years tend to stabilize.Pearson correlation analysis shows that the harvesting period of Scutellaria baicalensis G.was significantly correlated with the decay area,color value and chemical composition content.Conclusion Based on the comprehensive evaluation of harvesting age and chemical composition content,composition transformation,decay area and color change pattern,it is considered that Kuqin should be harvested within 3 years-7 years.
