摘要
目的探索怀地黄“菊花心”性状与化学成分之间相关性,为怀地黄辨状论质提供数据参考。方法取地黄断面,观察其形成层宽度、射线密度,Photoshop软件测定地黄中菊花心及形成层R(红)、G(绿)、B(蓝);CAD软件测定菊花心与韧皮部面积比值(以下均以心皮比表示);HPLC法测定梓醇、益母草苷、地黄苷D、毛蕊花糖苷、异毛蕊花糖苷、果糖、葡萄糖、棉子糖、蔗糖、水苏糖含量。采用SPSS 26.0软件对其“菊花心”性状与化学成分进行相关性分析。结果地黄形成层RGB均高于菊花心,北京3号RGB值最高,R、G、B值分别为196、185、85;地黄苷D、益母草苷与菊花心、形成层中RGB均值呈显著正相关,相关系数分别为0.515、0.523,与形成层宽度、射线密度呈极显著正相关,相关系数分别为0.887、0.887、0.904、0.904,其他化学成分与“菊花心”性状呈显著负相关或无显著相关性。结论所建立的怀地黄“菊花心”性状分析方法,可以全面概括通过直接观察得到的“菊花心”性状;地黄苷D、益母草苷与菊花心和形成层RGB均值、形成层宽度、射线密度呈相关性,是药材“辨状论质”的具体体现,可为地黄辨状论质提供理论支持。
Objective To provide reference for quality discrimination by character of Rehmannia glutinosa,through exploring the correlation between radial striations trait and chemical constituents of R.glutinosa.Methods Photoshop software was used to measure the color R(Red),G(Green),B(Blue)of radial striations and cambium.CAD software was used to measure the ratio of radial striations to phloem area(the following are expressed in carpel ratio).The contents of catalpol,leonuride,rehmaionoside D,acteoside,isoacteoside,fructose,glucose,raffinose,sucrose and stachyose were determined by HPLC.SPSS software was used to analysis the correlation between radial striations and the content of chemical composition.Results The RGB value of the cambium in R.glutinosa is higher than radial striations,and the RGB value of Beijing No.3 was the highest,with R,G,and B values of 196,185,and 85,respectively.Rehmaionoside D and leonuride were significantly positively correlated with RGB mean in the radial striations and cambium,with correlation coefficients of 0.515 and 0.523,respectively.In addition,Rehmaionoside D and leonuride were very significant positive correlation with the cambium width and ray density,with correlation coefficients of 0.887,0.887,0.904 and 0.904 respectively.Conclusion The method for analyzing the radial striations trait of R.glutinosa can comprehensively summarize the radial striations trait obtained by direct observation;Rehmaionoside D and leonuride were significantly positively correlated with RGB mean in the radial striations and cambium,and the cambium width and ray density,which is a concrete manifestation of the“quality discrimination by character”of the medicinal material,this result could provided theoretical support for the quality discrimination by character of R.glutinosa.
