有氧运动/高脂饮食干预父系C57BL/6小鼠对雄性子代小鼠脂肪组织PPAR-γ表达及其甲基化修饰的影响 被引量:1

Effects of Aerobic Exercise/Paternal High-Fat Diet Intervention on PPAR-γ and Its Methylation Modification in the Adipose Tissue of F1 Male Offsprings

摘要 目的:探究有氧运动/高脂饮食干预父系C57BL/6小鼠对雄性子代小鼠脂肪组织过氧化物酶体增殖物激活受体γ(PPAR-γ)表达及其甲基化修饰的影响。方法:同窝C57BL/6雄性小鼠(F0)自出生21天断乳后随机分为对照组、运动组和高脂组;运动组和高脂组分别进行6周有氧跑台训练及高脂饲料喂养。6周后将各组雄性小鼠与同窝雌鼠1︰1交配产下F1代小鼠。父系对照组的F1代小鼠为CO组,父系运动组的F1代为EO组,父系高脂组的F1代小鼠为HFO组。各组F1代小鼠4周龄时开始给予为期4周的高脂饮食干预。4周高脂饮食干预结束后检测F1代小鼠体重、体成分及血清相关指标;检测脂肪组织中PPAR-γ的表达及其甲基化水平。结果:(1)4周高脂饮食后,与CO组相比,HFO组小鼠体重、脂肪组织重量明显增加,血清中主要血脂指标显著上升;而EO组体重、脂肪组织重量明显减少,血清中主要血脂指标显著降低。(2)与CO组相比,EO组附睾脂肪组织中PPAR-γm RNA表达增加,而HFO组其表达降低。另EO组和HFO组附睾脂肪组织PPAR-γ启动子1区-437位点甲基化程度较CO组均升高。结论:有氧运动和高脂饮食干预父系小鼠可能通过改变雄性子代小鼠脂肪组织PPAR-γ启动子甲基化水平影响PPAR-γ的表达,从而对子代小鼠的代谢表型产生影响。 Objective To explore the effect of aerobic exercise/paternal high-fat diet intervention on C57 BL/6 mice on adipose tissue peroxisome proliferator-activated receptor-γ(PPAR-γ)expression and its methylation modification in F1 male offspring.Methods Male mice from the same litter of C57 BL/6 mice(F0)were randomly divided into a control,exercise and high-fat groups after weaning at 21 days of age.The exercise group and the high-fat group were trained for 6 weeks on the aerobic treadmill and fed high-fat diet respectively.Then male mice from each group were mated 1:1 with females from the same litter to give birth to F1 generation mice.The F1 generation mice in the control group were the CO group,the F1 generation in the exercise group were the EO group,and the F1 generation mice in the high-fat group were the HFO group.All F1 generation mice underwent a four-week highfat diet starting at 4 weeks of age.Body weight,and their body composition and serum-related indexes were measured after the intervention,and the expression of PPAR-γand its methylation level in the adipose tissue were evaluated.Results After 4 weeks of high-fat diet,compared with the CO group,the HFO group showed a significant increase in the average body weight and adipose tissue weight,but a significant increase in major serum lipid indexes,while the EO group showed a significant decrease in the average body weight and adipose tissue weight,but a significant decrease in major serum lipid indexes.Compared with the CO group,the average PPAR-γgene expression increased in the epididymal adipose tissue of the EO group,but decreased in the HFO group.The methylation of PPAR-γpromoter region 1,-437 site in the epididymal adipose tissue also increased in both EO and HFO groups compared to CO group.Conclusion Aerobic exercise and paternal high-fat diet intervention may affect PPAR-γexpression by altering the adipose tissue PPAR-γpromoter methylation level in the male offspring,thereby influencing the metabolic phenotype of offspring mice.
