真武汤治疗新型冠状病毒感染的网络药理学研究:基于五运六气理论 被引量:2

Network Pharmacology Study of Zhenwu Decoction in the Treatment of COVID-19 based on Five-Yun-Six-Qi Theory

摘要 目的:基于五运六气理论对真武汤干预新型冠状病毒(COVID-19)感染的机制进行网络药理学研究分析。方法:通过TCMSP数据库获取真武汤中附子、白术、茯苓、白芍、生姜的全部化学成分及靶点;通过Gencards数据库获取肺炎相关靶点,利用String平台进行蛋白质相互作用分析,构建PPI网络。采用David6.8对真武汤化学成分靶点与COVID-19肺炎靶点的交集的靶点进行GO富集分析,分析生物学过程;利用KEGG通路分析,分析代谢通路,并进行蛋白功能区域分析。采用Cytoscape3.7.2软件构建真武汤-成分-靶点-通路网络。结果:①真武汤中5种草药的作用靶点总计778个;COVID-19肺炎相关靶点4473个;草药靶点和COVID-19肺炎相关靶点的交集靶点47个,排序前3位的是:溶菌酶、孕激素受体、组织蛋白酶D;②真武汤含有300种不同化学分子成分;与47个交集靶点相关联的化学分子共计95种;排序前3位的分子为:醋酸、山柰酚、水杨酸;③GO富集分析出与47个交集靶点相关的生物学过程30个(P<0.01),排序前3位的是:序列特异性DNA结合转录因子活性的调控、MAPK活性的激活、过氧化氢分解代谢过程;KEGG通路富集分析得出靶点通路62个(P<0.01),排序前3位的是:2型糖尿病、NOD样受体信号通路、肿瘤坏死因子信号通路。SMART分析交集靶点主要作用蛋白功能区域有5个,排序前3位的是:ZnFC4、HOLI和STKc。结论:在五运六气理论指导下,选择真武汤治疗新型冠状病毒感染及相关肺炎具有理论基础与科学依据,对当前疫情防控具有一定指导价值。 Objective:To study the network pharmacologic mechanism of Zhenwu decoction in the treatment of COVID-19 based on the Five-Yun-Six-Qi theory.Methods:All chemical components and targets of Zingiber Officinale Roscoe、Paeoniae Radix Alba、Atractylodes Macrocephala Koidz.、Poria Cocos(Schw.)Wolf.、Aconiti Lateralis RadixPraeparatain in Zhenwu decoction were obtained by TCMSP database.Gencards database was used to obtain pneumonia-related targets,and String platform was used for protein interaction analysis to construct PPI network.David6.8 was used to conduct GO enrichment analysis on the targets at the intersection of the chemical composition target of Zhenwu decoction and the pneumonia target of COVID-19 to analyze the biological process.Metabolic pathway was analyzed by means of KEGG pathway analysis.The functional areas of the proteins were analyzed.Cytoscape 3.7.2 software was used to construct Zhenwu decoction-component-target-path network.Results:①The total number of action targets of 5 herbs in Zhenwu decoction was 778.There were 4473 targets associated with COVID-19 pneumonia.There were47 intersection targets of herbal and COVID 19 pneumonia-related targets,and the top 3 were Lysozyme、Progesterone receptor、Cathepsin D;②Zhenwu decoction contains 300 different chemical and molecular components;There are 95 chemical molecules associated with 47 intersection targets.The top three were acetic acid、kaempferol、salicylic acid;③GO enrichment was used to analyze 30 biological processes related to 47 intersections(P<0.01),the first three of which were regulation of sequence-specific DNA binding transcription factor activity、activation of MAPK activity、hydrogen peroxide catabolic process.The enrichment analysis of KEGG pathway led to 62 target pathways(P<0.01),the top 3 of which were Type II diabetes mellitus、NOD-like receptor signaling pathway、TNF signaling pathway.There are five main functional regions of the protein at the intersection of SMART analysis targets,and the first three are ZnF_C4,HOLI and S_TKc.Conclusion:The treatment of novel coronavirus infection and associated pneumonia with Zhenwu decoction under the guidance of the theory of Five-Yun-Six-Qi has theoretical basis and scientific basis.It has a certain value for the current epidemic prevention and control.
