摘要 目的:基于血清药物化学方法,对龙眼叶乙酸乙酯部位进行入血成分分析,探讨龙眼叶的药效物质基础。方法:采用UPLC-Q-TOF-MS/MS高分辨液质联用法,对乙酸乙酯部位及给药后大鼠血清样品进行分离后在负离子检测模式下进行总离子检测、二级碎片离子扫描,初步鉴定龙眼叶提取物在大鼠血中原型成分及代谢成分。结果:从含药血清中初步鉴定了共17个药源性成分,其中有1个原型成分曾从龙眼叶的提取物中得到分离,其余多为原型成分的苷元代谢产物。结论:初步确定了没食子酸乙酯、槲皮素、槲皮苷、木犀草素等成分可能为龙眼叶降血糖活性的物质基础。 Objective: To analyze the blood-transition constituents of ethyl acetate fraction of leaves from Dimocarpus longan based on the serum pharmacochemical method, and to investigate the pharmacological material basis of leaves from Dimocarpus longan.Methods: Using UPLC-Q-TOF-MS/MS high resolution liquid/mass spectrometry, the ethyl acetate fraction and serum samples of rats after administration were separated.Total ion detection and secondary fragment ion scanning were carried out under the negative ion detection mode to preliminarily identify the prototype components and metabolic components of extract from Dimocarpus longan leaves in the blood of rats.Results: A total of 17 drug-derived components were identified from the drug-containing serum, among which one prototype component was isolated from the extract of Dimocarpus longan leaves, and most of the others were aglycones metabolites of the prototype components.Conclusion: It is preliminarily determined that ethyl gallate, quercetin, quercitrin, luteolin and other components may be part of the substance basis of hypoglycemic activity of leaves of Dimocarpus longan.
