摘要
目的评价睡眠剥夺方法建立气虚血瘀证大鼠模型的可行性并构建系统性评价体系。方法24只SD大鼠随机分为正常对照组与模型组,每组12只。模型组采用每天随机14~16 h睡眠剥夺方法进行干预,连续6周;正常对照组大鼠不进行干预。比较两组大鼠的力竭游泳时间及体重系数,采用超声仪检测左心室射血分数(LVEF)、有效心输出量(CO)、心脏指数(CI)与每搏输出量(SV),采用旷场实验观察大鼠运动总路程、平均速度以及进入中央区域次数,并检测凝血功能、舌象的色彩分析及脉搏幅度,检测血浆/血清中促肾上腺皮质激素(ACTH)、烟碱型胆碱受体α1(chrnα1)、三磷酸腺苷(ATP)与二磷酸腺苷(ADP)含量以及心脏、血管病理结果。结果与正常对照组比较,模型组大鼠运动总路程、平均速度下降,进入中央区域次数增加,力竭游泳时间缩短,体重明显下降,血浆ACTH水平降低,血清chrnα1水平升高,心功能LVEF、CO、SV、CI均下降,血浆活化部分凝血酶时间(APTT)、凝血酶原时间(PT)降低,纤维蛋白原(FIB)增加,血清ATP含量降低,而ADP含量增加(P<0.05或P<0.01)。模型组大鼠舌象表现为红、红/绿、红/蓝、洋红色至绿色范围与黄色至蓝色范围值显著降低,脉搏幅度降低(P<0.05或P<0.01)。两组大鼠的心脏与血管组织病理未发生改变。结论睡眠剥夺方法可以成功建立气虚血瘀证大鼠模型,以主症、兼症、舌象、脉象为系统的评价体系对证候模型建立具有指导意义。
Objective To evaluate the feasibility of establishing a rat model of qi deficiency and blood stasis syndrome by sleep deprivation method and to construct a systematic evaluation system.Methods A total of 24 SD rats were randomly divided into a normal control group and a model group,with 12 rats in each group.The model group used the sleep deprivation method to intervene,and the intensity was randomized for 14 to 16 hours every day for 6 weeks;the normal control group rats did not intervene.Exhausted swimming time and body weight were compared between the 2 groups of rats.Left ventricular ejection fraction(LVEF),effective cardiac output(CO),cardiac index(CI),and stroke output(SV)were measured using an ultrasound system.Field experiments were used to observe the total distance of movement,average speed and the number of times of entering the central area.The blood coagulation function,tongue color analysis and pulse amplitude were tested,and the adrenocorticotropic hormone(ACTH)and nicotinic choline receptor,recombinant cholinergic receptor nicotinicα1(chrnα1),adenosine triphosphate(ATP)and adenosine diphosphate(ADP)content in plasma/serum,and cardiac and vascular pathological results were detected.Results Compared with the normal control group,the total distance of movement and average speed of the rats in the model group decreased,the number of times of entering the central area increased,the time of exhaustion swimming was shortened,the body weight was significantly reduced,the plasma ACTH level was reduced,the serum chrnα1 level was increased,and cardiac function LVEF,CO,SV,CI decreased,plasma activated partial thrombin time(APTT),prothrombin time(PT)decreased,fibrinogen(FIB)increased,serum ATP content decreased,and ADP content increased(P<0.05 or P<0.01).In the model group,tongue manifestations showed significant reductions in red,red/green,red/blue,magenta to green range,and yellow to blue range values,and pulse amplitudes were reduced(P<0.05 or P<0.01).The pathology of the heart and blood vessels of both groups of rats remained unchanged.Conclusion The sleep deprivation method can successfully establish a rat model of qi deficiency and blood stasis syndrome.The evaluation system based on the primary symptoms,companing symptoms,tongue,and pulse signs has a guiding significance for the establishment of syndrome models.
