白芷地上部分总香豆素的提取及其体外抗氧化活性考察

Extraction of Total Coumarins from the Above-Ground Parts of Angelica dahurica and Their in Vitro Antioxidant Activities

摘要 通过单因素试验结合正交试验,采用纤维素酶辅助提取白芷地上部分总香豆素,实验结果表明,总香豆素最佳提取工艺条件为纤维素酶浓度2.5 mg/mL、乙醇体积分数50%、提取温度55℃,提取时间60 min,此条件下的提取率为1.59%;对总香豆素进行体外抗氧化试验,结果显示,白芷地上部分总香豆素浓度1.2 mg/mL时,对DPPH自由基、羟基自由基和超氧阴离子自由基的清除率最大,分别为62.21%、74.55%、66.95%,说明白芷地上部分总香豆素具有一定的抗氧化活性。 Through single factor test combined with orthogonal test, cellulase-assisted extraction of total coumarins from the aboveground parts of Angelica dahurica was carried out. The experimental results showed that the optimal extraction conditions of total coumarins were cellulase concentration of 2.5 mg/mL, ethanol volume fraction of 50%, extraction temperature of 55˚C, and extraction time of 60 min. Under this condition, the extraction rate was 1.59%. The total coumarin was subjected to in vitro antioxidant test. The results showed that when the total coumarin concentration in the aboveground parts of Angelica dahurica was 1.2 mg/mL, the scavenging rates of DPPH free radical, hydroxyl radical and superoxide anion radical were the highest, which were 62.21%, 74.55% and 66.95%, respectively, indicating that the total coumarin in the aboveground part of Angelica dahurica had certain antioxidant activity.
