摘要
目的 检测食管癌患者外周血CD3+ T细胞分泌细胞因子的水平变化 ,分析Thl和Th2细胞的细胞因子免疫活动 ,为肿瘤的免疫治疗提供实验依据。方法 首先用刺激物刺激细胞 ,增加细胞内细胞因子的表达 ,然后加入荧光标记的特异性抗细胞因子单克隆抗体 ,特异性抗原抗体结合 ,最后应用流式细胞仪分析特异性细胞因子表达水平。同时用酶联免疫吸附法 (ELISA)检测相应的细胞因子。结果 食管癌患者Thl型细胞因子干扰素 -γ(IFN -γ)、白细胞介素 - 2 (IL - 2 )、白细胞介素 - 1 2 (IL - 1 2 )表达水平较正常对照组显著降低 ,Th2型细胞因子白细胞介素 - 4 (IL - 4 )和白细胞介素 - 1 0 (IL - 1 0 )表达水平较正常对照组升高 ,但差异无统计学意义。结论 食管癌患者Thl型反应模式处于弱势状态 ,相对地Th2型反应模式处于优势状态 ,Thl/Th2平衡失调 ,Thl/Th2平衡向Th2方向漂移。食管癌患者体内Th2型细胞因子模式占优势状态 ,这可能是肿瘤细胞发生免疫逃逸而避免被机体清除 ,从而导致肿瘤的发生或者转移的一种机制。
Objective To detect the level of Th1 and Th2 type cytokines of peripheral blood CD3+ T lymphocytes in the patients with esophageal cancer,and provide the laboratory evidence for the tumor immunotherapy. Methods The peripheral blood lymphocytes were stimulated by stimulator to enhance expression of cytokines. Specific fluorochrome-conjugated monoclonal antibody combined with antigen. The cytokines were detected by flow cytometry and ELISA. Results The levels of Th1 type cytokines such as interferon-γ(IFN-γ),interleukin-2(IL-2), interleukin-12(IL-12) in patients with esophageal cancer were significantly lower than those of normal controls. The level of Th2 type cytokines such as interleukin-4(IL-4), interleukin-10(IL-10) in patients with esophageal cancer were higher than those of normal controls, but not significant difference between them. Conclusions Th1/Th2 of T-lymphocytes is imbalance in esophageal cancer. Th1 type cytokines were inhibited, while Th2 type cytokines are relatively enhanced, so Th1/Th2 shift to Th2. It may be the mechanism of tumor arising and transferring by immune escaped from immunosurveillance.
