川芎嗪对同基因骨髓移植小鼠骨髓中干细胞因子表达的影响 被引量:1

Effects of Ligustrazine on Expression of Stem Cell Factor in Syngenic Bone Marrow Transplantation of Mice

摘要 目的 探讨川芎嗪促进骨髓移植后小鼠骨髓中干细胞因子表达的作用。方法 将小鼠随机分组 ,建立小鼠骨髓移植模型 ,对照组和川芎嗪组分别胃饲生理盐水 0 .2ml/只和川芎嗪注射液 2mg/只 ,2次 /天。在骨髓移植后的第 +7、+14、+2 1天 ,采用免疫组化的方法检测小鼠骨髓切片中干细胞因子的表达水平 ,记取外周血白细胞、血小板和骨髓有核细胞计数 ,并作骨髓组织学观察。结果 骨髓移植后第 +7、+14、+2 1天川芎嗪组干细胞因子的表达水平、外周血白细胞、血小板、骨髓有核细胞计数及骨髓细胞增生程度均高于骨髓移植的对照组 (P <0 .0 5或P <0 .0 1)。结论 川芎嗪能促进骨髓移植后骨髓中干细胞因子的表达 ,可能是川芎嗪重建造血的机制之一。 Objective To explore the effect of Ligustrazine on expression of stem cell factor in syngenic bone marrow transplantation(BMT)mice.Methods Mice were divides into three groups:normal control group,BMT control group,and Ligustrazine treated groups.BMT mouse models were established.The BMT control group and the ligustrazine treated group were given orally 0.2ml saline and 2mg Ligustrazine respectively twice a day.On the day 7,14,21 after BMT,mice were killed.Peripheral blood cell and bone marrow nucleared cell were detected.Histological observation of bone marrow was made and the stem cell factor expression was assayed by immunohistochemistry.Results In Ligustrazine treated group,the peripheral blood cell,bone marrow nucleared cell on the days 7,14,21after BMT were higher than the BMT control group. The expression than in the BMT control group(P<0.05 or P<0.01).Conclusion Ligustrazine can enhances SCF expression in bone marrow after syngenic bone marrow transplantation in mice,which may be related to the mechanisms underlying the Ligustrazine accelerating hematopoietic reconstitution in allogenic bone marrow transplantation.
