摘要
                
                    目的 :探讨碱烧伤后角膜内抗氧化酶 ,即超氧化物歧化酶 (SOD) ,过氧化氢酶 (CAT) ,谷胱甘肽过氧化酶 (GPx)的mRNA的表达的动态变化 ,从而进一步揭示角膜碱烧伤的损伤机制。方法 :根据Roper Hall分度法[1] ,分别制造轻、重两组碱烧伤模型 ,采用RT PCR的方法研究碱烧伤后大鼠角膜内抗氧化酶的mRNA表达的变化。结果 :在轻致伤组中 ,超氧化物歧化酶在伤后 2周时的mRNA表达下降与前期有明显差异 (P <0  0 5 )。过氧化氢酶在伤后 2天时其mRNA表达升至最高点 ,2周时明显下降。谷胱甘肽过氧化酶伤后 10分钟至 30分钟其mRNA的表达即为高水平。于 2天时 ,降至最低点。 1周后回升至正常。 2周时再次明显下降。在重致伤组中 ,超氧化物歧化酶在伤后 10分钟与烧伤后 2周其mRNA的表达明显下降 ,过氧化氢酶在伤后 10分钟就开始下降 ,30分钟至 6 0分钟达极低水平 ,2周时再次达低水平。谷胱甘肽过氧化酶在伤后 6 0分钟及2周时达最低水平。以上变化均具有显著性差异。结论 :角膜碱烧伤后 ,角膜内抗氧化酶的mRNA表达 ,并不是一味的下降 ,说明机体基因水平具有很高的应激能力。
                
                Objective:To observe the effect of alkali burns on the mRNA expression level of antioxidant related enzymes in rat corneas.Methods:Alkaline burns were made on rat corneas,according to Roper  Hall classification criteria,as mild and severe degree.The mRNA expression level of antioxidant related enzymes were determined with reverse transcription  polymerase chain reaction (RT  PCR).Results:The mRNA expression level of SOD decreased,that of CAT decreased very little and that of GPx increased significantly at 10 minutes after alkaline injury.However,at 60 minutes~2 days,the mRNA expression levels of each antioxidant related enzymes were at high levels,but decreased again at 2 weeks.Conclusion:This study suggests that the tissues have high ability to control stimulate at gene level.Clinically,this may be helpful in treating corneal alkaline injury.
