摘要
目的研究苦参-蛇床子药对提取物(extract of sophorae flavescentis radix-Cnidii fructus Coupletmedicines,ESCC)对白念珠菌外阴阴道念珠菌病(VVC)临床株的凋亡诱导作用。方法以256、512、1 024μg·mL-1的ESCC处理白念珠菌VVC临床株后,以扫描电子显微镜观察白念珠菌生物膜的形态结构;采用流式细胞术检测白念珠菌生物膜细胞凋亡率、活性氧(reactive oxygen species,ROS)和线粒体膜电位(mitochondrial membrane potential,MMP);采用荧光显微镜观察白念珠菌生物膜细胞Caspase酶活性和细胞核的损伤情况。结果终浓度>256μg·mL-1的ESCC可影响白念珠菌VVC临床株生物膜细胞的形态结构,提高胞内ROS,降低MMP,升高Caspase酶活性,损伤细胞核。结论 ESCC可通过诱导白念珠菌VVC临床株生物膜细胞凋亡而发挥抗真菌作用。
Objective This study aimed to investigate the apoptosis induction activity of extract of Sophorae flavescentis radix-Cnidii fructus couplet medicine(ESCC)on biofilms formed by Candida albicans VVC isolates.Methods Scanning electron microscopy(SEM)was used to observe the biofilms formed by C.albicans.The effect of ESCC on reactive oxygen species(ROS),mitochodria membrane potential(MMP)and apoptosis rate of C.albicans were detected by flow cytometry.Caspase activity and nuclear fragmentation were observed by inverted microscopy.Results The results showed that ESCC with a concentration higher than 256μg·mL-1 could increase the apoptosis ratio,intracellular ROS and Caspase activity in C.albicans,meanwhile decrease MMP and induce nuclear condensation and fragmentation.Conclusion This study demonstrated that ESCC had prominent apoptosis induction activity which exerting significant antifungal effect on C.albicans.
