扶正解毒化瘀颗粒对多器官损伤小鼠肺组织的保护作用 被引量:1

Protective Effect of Fuzheng Jiedu Huayu Granule on Lung Tissue of Multi-organ Injury Induced in Mouse

摘要 目的探讨扶正解毒化瘀颗粒对克雷伯杆菌肺炎所致多器官损伤小鼠肺组织的保护作用机制。方法将128只小鼠随机分为对照组、模型组、西药组和中药组,每组32只;除对照组外,其余各组采用气管插管法直接注入肺炎克雷伯杆菌制备多器官损伤模型;造模前3天,中药组给予扶正解毒化瘀颗粒混悬液[0.01 mL/(g·d)]、西药组给予左氧氟沙星胶囊混悬液[0.01 mL/(g·d)]进行灌胃给药,对照组和模型组给予等量生理盐水灌胃,分别于造模后第1、3、5、7天每组各取8只小鼠,取出肺脏;左肺进行组织匀浆,离心后取上清液,ELIs A法检查不同感染时相小鼠肺组织中细胞因子TNF-α、IL-1β、IL-6、IFN-γ、IL-10水平;右肺HE染色,光学显微镜观察不同感染时相小鼠肺组织病理变化。结果与对照组比较,模型组小鼠在感染后第1天、第3天肺组织中TNF-α、IL-1β、IL-6、IFN-γ、IL-10等细胞因子水平显著升高(p <0.05或p <0.01),肺组织病理损伤明显;与模型组比较,中药组小鼠在感染后第1天肺组织中TNF-α、IL-1β、IL-6、IFN-γ等促炎细胞因子水平显著降低(p <0.05),抗炎细胞因子IL-10水平显著增高(p <0.05),肺组织病理损伤减轻。结论扶正解毒化瘀颗粒对克雷伯杆菌肺炎所致多器官损伤小鼠肺组织具有保护作用,其机制可能与调节促炎/抗炎细胞因子网络平衡有关。 Objective To reveal the possible mechanism of Fuzheng Jiedu Huayu granule on lung tissue protection of multiple organ injury induced by Klebsiella pneumoniae,through cytokines and myocardial histopathology.Methods 128 mice were randomly divided into control group,model group,western medicine group and traditional Chinese medicine group with 32 animals in each group;except the control group,the other groups were directly injected into Klebsiella pneumoniae by tracheal intubation to prepare a multi-organ dysfunction model;3 days before modeling,the Chinese medicine group was given Fuzheng Jiedu Huayu granule suspension[0.01 mL/(g·d)],western medicine group was given levofloxacin capsule suspension[0.01 mL/(g·d)]for intragastric administration,and the control group and model group were given the same amount of normal saline,8 mice were sacrificed on the 1 st,3 rd,5 th,and 7 th days after modeling respectively;the left lung tissue was homogenized and the levels of TNF-α,IL-1β,IL-6,IFN-γ,IL-10 were detected in different infection phases by ELISA;right lung tissue was HE staining,and the pathological changes of different infection phases were observed under light microscope.Results Compared with the control group,the cytokines level of TNF-α,IL-1β,IL-6,IFN-γ,IL-10 in the lung tissue of the model group were significantly increased on the 1 st and 3 rd day after infection(P<0.05 or P<0.01),and the pathological damage of lung tissue was obvious.Compared with the model group,on the 1 st day after infection,the level of pro-inflammatory cytokines such as TNF-α,IL-1β,IL-6 and IFN-γin lung tissue were significantly decreased(P<0.05),and the level of anti-inflammatory cytokine IL-10 was obviously increased(P<0.05)in the traditional Chinese medicine group,and the pathological damage of lung tissue was reduction.Conclusion Fuzheng Jiedu Huayu granule has protective effect on lung tissue of mice with multiple organ function damage caused by Klebsiella pneumoniae.The mechanism may be related to reducing the expression of pro-inflammatory cytokines such as TNF-α,IL-1,IL-6 and IFN-γin lung tissue,increasing the expression of anti-inflammatory cytokine IL-10,and achieving the balance of pro-inflammatory anti-inflammatory cytokine networks.
