摘要
目的 :观察成年猫一侧坐骨神经切断后Fas、FasL和Caspase - 3在脊髓的分布。方法 :将成年猫坐骨神经切断后 2 4小时 ,处死取其脊髓 (L4)制作 8μm厚石蜡切片 ,用Fas、FasL、Caspase - 3(效价 :1:2 5 0 ,1:2 5 0 ,1:2 5 0 ,)行免疫组织化学ABC法染色。观察Fas、FasL、Caspase - 3在脊髓中的分布以及亚细胞定位。结果 :Fas阳性神经元遍布整个灰质 ,以背角为主 ,胞浆染色 ,灰质内胶质细胞阳性反应 ,细胞核染色。FasL阳性神经元遍布整个灰质 ,胞浆染色 ,未见胶质细胞阳性染色。Caspase - 3阳性反应神经元胞浆染色。结论 :一侧坐骨神经切断后 ,有Fas、FasL和Caspase- 3在猫脊髓分布 。
Objective:To observe the Fas,FasL and Caspase-3 are distributing in spinal cord of adulthood cat following unilaterals sciatic nerve transection.Method:Take the spinal cord(L4)are made 8 μm thick to cut into paraffin slices,use the Fas,FasL,Caspase-3 rabbit antiserum(1:250,1:250,1:250)by the immunohistochemistry ABC method.Observe the Fas,FasL,Caspase-3 are distributing in the spinal cord of and the location of second cell.Result:The Fas immunoreactive positive produceare distribute in the cytoplasm of neurons,the neurons of dorsal horn of gray matter stain obviously than other,there are no positive neuoglia cells in the whiter and gray matter;The FasL immunoreactive positive produces are distribute in the cytoplasm of neurons of gray matter too.Caspase-3 are in the cytoplasm of neurons in the spinal cord of cat.Conclusion:The Fas,FasL and Caspase-3 distribute in the spinal cord of cat following unilaterals sciatic nerve transection and hint these factors relevant with the function of apoptotic signal transmitter in spinal cord.
