摘要
目的:探讨银杏叶提取物(EGb)对大鼠脊髓全横断损伤后神经生长因子(NGF)及脑源性神经营养因子(BDNF)蛋白水平表达变化的影响。方法:T10水平横断成年SD大鼠脊髓,取尾端制作冰冻切片,采用免疫组化ABC法染色。观察并计数脊髓腹角NGF、BDNF的表达变化。结果:在EGb组,伤后3 d NGF及BDNF的表达即开始增高,第7天时更明显,然后开始下降,但直到21d仍高于假手术组(P〈0.05);与单纯脊髓横断组及生理盐水组在3d、7d比较,均明显增加(P〈0.05)。结论:提示EGb可通过增加NGF、BDNF表达而促进脊髓损伤早期修复。
Objective To explore the effect of extract of ginkgo bilobae ]eaves (EGb) on the expression of NGF and BDNF in spinal cord following transected injury. Methods Transected spinal cord injury was performed at T10 in adult SD rats. The L1 ~ 2 spinal segments were removed out and made into 20μm frozen sections. Dynamic changes of NGF and BDNF expression of spinal cord were observed at different time with ABC method of immunohistochemistry. Results The increased expression of NGF and BDNF were observed at 3d after operation and reached a peak at 7d in EGb group. The expression of NGF and BDNF were down - regulated after 7d, but still higher than those in sham group until 21d ( P 〈0.05 ). The number of NGF and BDNF positive neurons in the ventral horn of spinal cord in EGb group increased more than that in transected spinal cord injury group and normal sodium group at 3d and 7d ( P 〈0. 05 ). Conclusion These data give a clue to elucidate that EGb is beneficial to the early repair of spinal cord injury.
