摘要
从我所保藏的菌种中筛选并经紫外线处理获得一株产细菌溶解酶的侧孢芽孢杆菌。研究了产生溶解酶的最适条件;培养基中葡萄糖的最适浓度为0(?)2%,超过此浓度抑制此酶的产生;在菌的稳定生长期,酶的产量最高。用 DEAE-纤维素柱层析提纯的酶制品,在聚丙烯酰胺凝胶电泳中呈现一条主带。培养滤液和酶制品能溶解全部供试溶菌谱的革兰氏阳性菌,对个别几株铜绿色假单胞菌也有较低的溶解活性。溶菌的最适 pH 是7.5,最适温度是45℃,Tris-HCl 缓冲液的浓度超过0.05M,溶菌活性就突然下降。
A bacterial lytic enzyme producingstrain of B.laterosporus was screened outfrom the collection of this Institute andfurther improved by U.V.treatment.Aglucose concentration of 0.2% is mostsuitable for enzyme production and abovethis value has a repression effect.Theproduction of the enzyme reaches itsmaximum at stationary growth phase.The enzyme was purified by DEAE-cellulose chromatography and showed onemajor band in polyacrylamide gel electro-phoresis.The enzyme lyses the Grampositive bacteria tested and shows a de-tectable lytic activity toward severalstrains of Pseudomonas aeruginosa.Theoptimum pH is 7.5 and the optimum tem-perature is 45℃ for lysis.The lyticactivity decreases when the Tris-HClbuffer concentration exceeds 0.05M.
