反相高效液相色谱法同时测定6种氟喹诺酮类药物 被引量:19

摘要 建立了一种反相高效液相色谱 荧光检测法同时测定血浆中6种氟喹诺酮类药物(FQS)的方法。考察了6种FQS的保留值与流动相组成及pH值的关系,优化了色谱条件及样品前处理方法。确定了以ChiraDex为色谱柱、乙腈 甲醇 Britton Robinson缓冲液(体积比为73∶7∶20,pH5 7)为流动相的最佳条件。该法用于血浆样品中FQS的测定,其回收率高于99 0%。该法简便、快速、准确、灵敏度高、重现性好。 A high performance liquid chromatographic method using fluorescence detection was developed for simultaneous separation and determination of 6 fluoroquinolones. The effects of mobile phase composition and pH value on retention parameters of 6 fluoroquinolones were investigated. Chromatographic conditions and sample pretreatment methods were optimized. The column used was a Chira Dex column (5 μm, 250 mm×46 mm id) with acetonitrilemethanolBrittonRobinson buffer solution (73∶7∶20, v/v) as mobile phase , which was adjusted using BrittonRobinson buffer solution (004 mol/L equally for phosphoric acid, acetic acid and boric acid). It was shown that pH 57 gave the best separation. The excitation and emission wavelengths were 283 nm and 448 nm, respectively. The flow rate was 06 mL/min. The method was applied to the determination of 6 fluoroquinolones in human plasma. The recoveries were more than 990%. The method is simple, rapid, accurate, sensitive and reproducible.
