海芦笋内生真菌基于PKS Ⅰ型功能基因的分离与鉴定 被引量:3

摘要 以采自新疆盐湖的野生海芦笋为研究对象,以聚酮合成酶(polyketide synthase,PKS)Ⅰ型功能基因为指标,采用平板划线技术、斜面分离纯化技术分离内生真菌,从中筛选出1株含有PKSⅠ型基因、编号为Salix1的内生真菌,进而PCR扩增该菌的18S rDNA和ITS1-5.8S-ITS2 rDNA目标序列,在美国国立生物技术信息中心(National Center of Biotechnology Information,NCBI)上进行同源性分析,构建系统发育树,结合形态学观察,将菌株Salix1鉴定为杂色曲霉。 In this study, a fungal endophyte with type Ⅰ polyketide synthase(PKS) named as Salix 1 was isolated from wild Salicornia bigelovii collected from a salt lake in Xinjiang by streak plate and slant culture methods. The 18S rDNA and ITS1-5.8S-ITS2 rDNA sequences from the strain were amplified by PCR and sequenced for sequence homology analysis using NCBI BLAST, and phylogenetic trees were constructed. Based on phylogenetic analysis combined with morphological observation, the strain Salix 1 was identified as Aspergillus versicolor.
