摘要
采用单因素试验研究时间、温度和料液比对番茄红素提取率的影响。在单因素试验的基础上,用L9(34)正交试验,寻找影响番茄红素提取率的最佳水平组合。试验在440~510 nm范围内测量番茄红素的吸光度,以确定番茄红素的最大吸收波长,在番茄红素的最大吸收波长下,比较不同种类的有机溶剂的提取效果,寻找提取效果较好的溶剂。然后绘制标准曲线。根据单因素试验和正交试验结果计算番茄红素的含量。试验结果显示,番茄红素的最大吸收波长是475 nm,利用乙酸乙酯作为提取溶剂时,提取效果最好。单因素试验结果显示,番茄红素的提取率随着提取时间、温度和料液比的增加而增加。当时间、温度和料液比分别达到70 min、45℃和1∶3后,提取率不再上升。正交试验显示,番茄红素提取的最优组合是A2B1C2。综合单因素和正交试验结果,用乙酸乙酯作溶剂时,影响番茄红素提取的各因素的最佳水平组合为:提取时间70 min、提取温度40℃、料液比1∶3。
The experiment studied the effect of time,temperature and solid-liquid ratio on extraction of lycopene.The purpose of the experiment was to complete the orthogonal test based on L9(34)on the basis of single factor test,and to determine the optimal extractionprocess of lycopene.In the experiment,the absorbance of fresh tomato extract was measured in the range of 440 to 510 nm to determine the maximum absorption wavelength of lycopene.The results of different types of organic solvents were compared at the determined maximum absorption wavelength to choose the optimum extraction solvent.Concentration of lycopene was calculated according to the standard curve.The results showed that the maximum absorption wavelength of lycopene was 475 nm,and the extraction effect was best when ethyl acetate was used as the extraction solvent.Data of single factor experiment showed that the extraction rate of lycopene increased with the increase of extraction time,temperature and solid-liquid ratio.When the time,temperature and solid-liquid ratio reached 70 min,45℃and 1∶3(g∶m L),the extraction rate stopped rising.The best combination for extracting lycopene obtained by using L9(34)orthogonal experiment was A2 B1 C2.In conclusion,based on all the results of present study,the optimal combinationwasasfollows:extractiontimeof 70 min,extractiontemperatureof40℃,solid-liquidratio of 1∶3(g∶m L).
