胃肠安对人胃癌MGC-803裸鼠皮下移植瘤中乏氧相关蛋白表达的影响

摘要 目的:观察MGC-803人胃癌裸鼠皮下移植瘤模型经胃肠安干预后对其相关乏氧蛋白表达的影响,探讨胃肠安对MGC-803人胃癌转移的作用机制。方法:选用BALB/C裸鼠,以人胃癌MGC-803细胞在其皮下注射建立移植瘤模型,随机分为3组并给予相应干预,分别为胃肠安组(胃肠安煎剂0.3 ml灌胃,1次/d)、5-氟尿嘧啶(5-Fu)组(5-Fu 1 mg腹腔注射,1次/周)、模型组(0.9%Na Cl溶液0.3 ml灌胃,1次/d),干预时间均为8周。用免疫组化法检测乏氧相关蛋白包括乏氧诱导因子-1α(HIF-1α)、转化生长因子-β1(TGF-β1)、赖氨酰氧化酶(LOX)、类赖氨酰氧化酶2(LOXL2)、结缔组织生长因子(CTGF)的表达。结果:胃肠安组裸鼠胃癌组织乏氧相关蛋白HIF-1α、TGF-β1、LOX、LOXL2、CTGF表达均较模型组明显降低,两组比较差异均有统计学意义(P<0.01)。结论:胃肠安能明显降低人胃癌MGC-803裸鼠皮下移植瘤中HIF-1α、TGF-β1、LOX、LOXL2、CTGF的表达,提示胃肠安可能通过下调乏氧相关蛋白的表达而起到抑制肿瘤转移的的作用。 Objective: To observe the effects of Weichang'an on hypoxia related proteins in nude mice with subcutaneous xenograft tumor of human gastric carcinoma MGC-803,and discuss the mechanism of Weichang'an on MGC-803 human gastric cancer metastasis.Methods: BALB/C nude mice were selected and the xenograft tumor model was established with human gastric carcinoma MGC-803 cells by subcutaneous injection. The model nude mice were randomly divided into the Weichang'an group,5-Fluorouracil( 5-Fu) group and model group. The Weichang'an group was treated with Weichang'an decoction at volume of 0.3 ml by intragastric administration once a day,the 5-Fu group was treated with 5-Fu at dose of 1 mg by intraperitoneal injection once a week,and the model group was treated with0.9% Na Cl solution at volume of 0. 3 ml by intragastric administration once a day. All the treatments were performed for 8 weeks. The expressions of hypoxia related proteins were detected by immunohistochemistry,including hypoxia inducible factor-1α( HIF-1α),transforming growth factor β1( TGF-β1),lysyl oxidase( LOX),lysyl oxidase-like 2( LOXL2) and connective tissue growth factor( CTGF).Results: Compared with the model group,the expressions of HIF-1α,TGF-β1,LOX,LOXL2 and CTGF in gastric cancer tissue of nude mice were obviously decreased in the Weichang'an group,with statistically significant differences( P< 0.01). Conclusion: Weichang'an can obviously down-regulate the expressions of HIF-1α,TGF-β1,LOX,LOXL2 and CTGF in human gastric carcinoma MGC-803 subcutaneous transplanted tumor in nude mice,which suggests that Weichang'an may inhibit the tumor metastasis through the downregulation of hypoxia related proteins expression.
