摘要
目的观察创伤后应激障碍(PTSD)患者外周血单个核细胞p11 mRNA表达变化,并探讨其意义。方法PTSD患者19例(PTSD组),经历创伤事件但未发生PTSD的患者13例(非PTSD组),健康体检者19例(对照组)。采用实时荧光定量聚合酶链式反应检测各组p11 mRNA,以创伤后应激障碍量表平民版(PCL-C)、汉密顿焦虑量表(HAMA)、汉密顿抑郁量表(HAMD)评估患者病情,Pearson相关分析法分析p11 mRNA表达与HAMA、HAMD、PCL-C评分的相关性。结果 PTSD组、非PTSD组、对照组p11 mRNA相对表达量分别为-0.495±0.920、0.911±1.474、-0.000±0.791,PTSD组、对照组分别与非PTSD组比较,P均<0.05。PTSD组HAMA、HAMD、PCL-C评分分别为(21.63±7.79)、(25.11±8.56)、(55.84±6.35)分,非PTSD组分别为(10.31±6.22)、(13.00±9.88)、(31.77±5.09)分,两两比较,P均<0.05。p11 mRNA相对表达量与HAMD评分呈负相关(r=-0.299,P=0.033),与PCL-C评分呈负相关(r=-0.385,P=0.030),与HAMA评分无相关性(r=-0.233,P=0.111)。结论PTSD患者p11 mRNA表达降低,检测此指标有助于鉴别诊断PTSD与非PTSD。
Objective To investigate the expression changes and significance of p11 mRNA in the peripheral blood mononuclear cells( PBMCs) of patients with post-traumatic stress disorder( PTSD). Methods In the PTSD group,there were 19 cases of patients; during the 19 patients,13 cases of patients( who were taken as the non-PTSD group) did not developed into PTSD; another 19 cases of healthy people were selected as the control group. The mRNA expression levels of p11 of all participants were estimated by using real-time fluorescence quantitative polymerase chain reaction. Post-traumatic stress disorder check-list-civilian version( PCL-C),Hamilton Anxiety Scale( HAMA) and Hamilton Depression Scale( HAMD) were used to access the condition of patients. Pearson correlation analysis was used to analyze the correlations of p11 mRNA expression with HAMA,HAMD and PCL-C scores. Results The relative expression of p11 mRNA in the PTSD group,non-PTSD group and control group was- 0. 495 ± 0. 920,0. 911 ± 1. 474 and- 0. 000 ± 0. 791,respectively. The PTSD group and control group showed a significant difference as compared with the non-PTSD group( all P <0. 05). The scores of HAMA,HAMD and PCL-C in the PTSD group were( 21. 63 ± 7. 79),( 25. 11 ± 8. 56) and( 55. 84± 6. 35),respectively. In the non-PTSD group,the scores were( 10. 31 ± 6. 22),( 13. 00 ± 9. 88) and( 31. 77 ± 5. 09),respectively. Multiple comparison showed there were significant differences between these two groups( all P < 0. 05). In the PBMCs,the relative expression of p11 mRNA was negatively correlated with HAMD( r =- 0. 299,P = 0. 033),was negatively correlated with PCL-C( r =- 0. 385,P = 0. 030),and was not related with HAMA( r =- 0. 233,P = 0. 111).Conclusion The p11 mRNA expression level is decreased,which may help in the diagnosis and differential diagnosis of PTSD and non-PTSD.
