摘要
本实验研究了应用不同的乳化剂、种子培养周期、接种量、发酵周期及溶氧量对耻垢分枝杆菌Mycobacterium smegmatis ZS3进行发酵生产4-雄烯二酮的影响,并对从摇瓶发酵到200 L发酵罐发酵的效价及进行200 L发酵罐菌种发酵的流加补糖方案进行了研究。研究结果显示,在进行200 L发酵罐菌种发酵的过程中,将溶氧量控制为25%-30%,在发酵60 h后进行流加补葡萄糖且将残糖的浓度维持在1 g/L时,最终植物甾醇转化为4-雄烯二酮的效价高于9.5 g/L。
The effect of emulsifiers, cultivation cycles of seed, inoculum concentration, dissolved oxygen and other parameters of 4-androstendione in shaking flask experiments, as well as with continuous feeding process in 200 L fermentation were studied. Results showed that by controlling the dissolved oxygen content between 25% to 30% in a 120-hours bath fermentation, the titer of 4-androstendione in 200 L fermenter reached above 9.5 g/L; while using continuous feeding process with glucose and keeping the concentration of glucose 1 g/L.
