摘要
根据黄体的大小和形态 ,判断屠宰母牛的发情时期 ,分别收集发情第 3天 (D3)和第 7天 (D7)的输卵管液 (OF)和子宫液 (UF) ,通过HPLC分析游离氨基酸种类及含量。总共检测到 2 3种氨基酸 ,包括除Cys外的 19种必需和非必需氨基酸 ,以及另外四种 (βAla、Tau、Orn和Cit)非蛋白质氨基酸。OFD3、UFD3、OFD7和UFD7的游离氨基酸总量分别为 31 6、 46 9、 2 6 3和 17 2mmol/L ,其中 ,Gly含量最高 ,分别为14 7、14 4、11 1和 4 4,其次为Glu、Gln和Ala ,其他氨基酸的含量均接近或低于 1mmol/L。结果表明 :氨基酸总量及个别氨基酸含量在不同体液及不同发育时期均存在一定程度的差异 [动物学报 49(1) :80~ 85 ,2 0 0 3]。
It is common practice to add amino acids to an embryo culture medium from a commercial stock solution originally designed for cell growth in culture.This study was conducted to measure amino acid levels in bovine oviductal fluid(OF)and uterine fluid(UF)at different preimplantation stages by HPLC.The stage of the estrous cycle was estimated according to morphological characteristics of the corpus luteum.A total of 23 free amino acids were identified, including 19 kinds of protein(Cys excluded)and another 4 kinds of non-protein amino acids(βAla、Tau、Orn and Cit).The total content of amino acids on day 3 OF, day 3 UF,day 7 OF and day 7 UF were 31.6,46.9,26.3 and 17.2 mmol/L, respectively.Concentrations of Gly in the above four tested fluids were the highest; 14.7、14.4、11.1 and 4.4 mmol/L, respectively.Concentrations of Glu, Ala and Gln were lower than Gly, but higher than the other amino acids which had concentrations close to or lower than 1.0 mmol/L.These results indicate that concentrations of individual and total amino acids varied in different fluids at different stages of estrous cycle [Acta Zoologica Sinica 49(1):80-85, 2003].
