摘要
[目的]探究壮药血力除疲汤对慢性疲劳综合征(CFS)大鼠行为学及下丘脑-垂体-肾上腺轴激素[皮质酮(CORT)、促肾上腺皮质激素(ACTH)]水平的影响。[方法]将72只大鼠随机分为6组:正常组、模型组、血力除疲汤低剂量组、血力除疲汤中剂量组、血力除疲汤高剂量组、逍遥散组,每组12只。以冷水游泳联合慢性束缚法建立CFS模型,造模后进行药物干预,血力除疲汤低、中、高剂量组大鼠分别灌胃给予6 g/kg、12 g/kg、24 g/kg的血力除疲汤,逍遥散组灌胃给予0.06 g/kg的逍遥散,正常组、模型组灌胃给予纯净水,灌胃体积均为10 ml/kg,每天1次,连续干预4周。以力竭游泳、悬尾与旷场实验检测大鼠行为学变化,测定大鼠下丘脑、垂体与肾上腺指数;采用ELISA法检测血清CORT、ACTH、白细胞介素6(IL-6)、肿瘤坏死因子α(TNF-α)的水平;以HE染色进行大鼠股四头肌病理形态检测;以流式细胞实验检测各组大鼠脑细胞凋亡情况。[结果]与正常组比较,模型组大鼠力竭游泳时间缩短,站立次数及水平跨格数减少,血清CORT、ACTH水平降低(P<0.05),悬尾不动时间延长,下丘脑、垂体、肾上腺指数,IL-6、TNF-α水平及脑细胞凋亡率升高(P<0.05);与模型组比较,血力除疲汤低、中、高剂量组及逍遥散组大鼠力竭游泳时间延长,站立次数及水平跨格数增加,血清CORT、ACTH水平升高,悬尾不动时间缩短,下丘脑、垂体、肾上腺指数,IL-6、TNF-α水平及脑细胞凋亡率均降低(P<0.05),且血力除疲汤各剂量组间呈剂量依赖性(P<0.05);血力除疲汤高剂量组大鼠各观察指标相比逍遥散组无明显差异(P>0.05)。与模型组大鼠比较,血力除疲汤各剂量组大鼠股四头肌形态和炎性细胞浸润情况显著改善。[结论]壮药血力除疲汤可改善CFS大鼠行为学,提高CORT及ACTH水平,降低IL-6、TNF-α水平及脑细胞凋亡率,改善股四头肌炎性损伤。
[Objective]To investigate the effects of Zhuang materia medica Xueli Chupi decoction on behavioral performance and hypothalamic-pituitary-adrenal axis hormones[corticosterone(CORT)and adrenocorticotropic hormone(ACTH)]in rats with chronic fatigue syndrome(CFS).[Methods]Seventy-two rats were randomly divided into six groups(n=12 per group):normal group,model group,low-dose Xueli Chupi decoction group,medium-dose Xueli Chupi decoction group,high-dose Xueli Chupi decoction group,and Xiaoyao San group.The CFS model was established using a combination of cold-water swimming and chronic restraint stress.After modeling,drug interventions were administrated with:the low-,medium-,and high-dose Xueli Chupi decoction groups received intragastric administration of the decoction at 6 g/kg,12 g/kg,and 24 g/kg,respectively;the Xiaoyao San group received 0.06 g/kg of Xiaoyao San;the normal and model groups received purified water.The administration volume was 10 ml/kg once daily for four consecutive weeks.Behavioral changes were assessed using the exhaustive swimming test,tail suspension test,and open field test.The organ indices of the hypothalamus,pituitary,and adrenal glands were measured.Serum levels of CORT,ACTH,interleukin-6(IL-6),and tumor necrosis factor-α(TNF-α)were detected by ELISA.Pathomorphological changes in the quadriceps muscle were observed via HE staining.Apoptosis of brain cells was detected by flow cytometry.[Results]Compared with the normal group,the model group showed significantly shortened exhaustive swimming time,reduced number of standing episodes and horizontal grid crossings,decreased serum levels of CORT and ACTH(P<0.05),prolonged immobility time in the tail suspension test,and increased hypothalamus,pituitary,and adrenal indices,IL-6 and TNF-α levels,as well as brain cell apoptosis rate(P<0.05).Compared with the model group,all Xueli Chupi decoction dose groups and the Xiaoyao San group exhibited prolonged exhaustive swimming time,increased number of standing episodes and horizontal grid crossings,elevated serum CORT and ACTH levels,shortened immobility time in the tail suspension test,and reduced hypothalamus,pituitary,and adrenal indices,IL-6 and TNF-α levels,and brain cell apoptosis rate(P<0.05).A dose-dependent effect was observed among the different dose groups of Xueli Chupi decoction(P<0.05).No significant differences were found in any observed indicators between the high-dose Xueli Chupi decoction group and the Xiaoyao San group(P>0.05).Compared with the model group,all Xueli Chupi decoction dose groups showed significant improvements in quadriceps morphology and inflammatory cell infiltration.[Conclusion]Zhuang materia medica Xueli Chupi decoction can improve behavioral performance,increase CORT and ACTH levels,reduce IL-6 and TNF-α levels and brain cell apoptosis rate,and ameliorate inflammatory damage in the quadriceps muscle in CFS rats.
