秋冬季马尾松穗条生理生化特性研究

摘要 比较秋冬季马尾松穗条的内源激素、营养物质、酶的含量和扦插生根效果,为马尾松扦插苗在生产中的应用提供理论依据。以马尾松当年生半木质化枝条为材料,采用液相色谱-串联质谱联用法(LC/MS/MS)检测内源激素,标准方法测定可溶性糖、可溶性蛋白质、总氮含量以及超氧化物歧化酶(SOD)、过氧化氢酶(CAT)、多酚氧化酶(PPO)、过氧化物酶(POD)、吲哚乙酸氧化酶(IAAO)等5种相关酶的活性,同时进行扦插。结果显示:GA的含量为0.023~0.147 ug·g^(-1),ZT的含量为0.045~0.114 ug·g^(-1),IAA含量为0.03~0.094 ug·g^(-1),ABA含量为0.143~0.492 ug·g^(-1),IAA∶ABA比值为0.12~0.42;可溶性糖、可溶性蛋白质、总氮含量和C/N均在1月份最高,分别为6.63%、6.78 mg·g^(-1)、30.51 mg·g^(-1)和2.17;SOD为49.52~424.4 U·mg^(-1),POD为25.49~84.61 U·(g.min)^(-1),PPO为29.77~77.94 U·(g.min)^(-1),CAT为198.23~342.94 g FW·min,IAAO为40.22~137.2 ug IAA·(h·ml)^(-1);生根率11月最高,为81.7%,其次是12月,为81.2%。生产中,在保证温度和湿度的条件下,可以在11月和12月采穗扦插,以获得马尾松无性繁殖植株。 Masson pine(Pinus massoniana)is one of the typical tree species in China.This study aims to compare the content of endogenous hormones,nutrients,enzymes and rooting effects of Masson pine cuttings in autumn and winter.Among them,the cutting is a small shoot cut from the tree,and then placed in soil to form the new one.The raw materials were also taken as the semi-lignified branches in the current year.Liquid chromatographyandem mass spectrometry(LC/MS/MS)was used to detect the endogenous hormones.A systematic investigation after cuttings was made to determine the contents of soluble,soluble protein,total nitrogen,and the activities of five related enzymes,including the superoxide dismutase(SOD),catalase(CAT),polyphenol oxidase(PPO),peroxidase(POD),and indole acetic acid oxidase(IAAO).The results showed that the content of GA was 0.023~0.147 ug·g^(-1);the content of ZT was 0045~0.114 ug·g^(-1),the content of IAA was 0.03~0.094 ug·g^(-1),the of ABA was 0.143~0.492 ug·g^(-1),and the ratio of IAA/ABA was 0.1~0.42;Meanwhile,the highest contents of soluble sugar,soluble protein,total nitrogen,and C/N were 6.6%,6.78mg·g^(-1),30.51mg·g^(-1),and 2.17,respectively,in January;Specifically,the SOD,POD,PPO,CAT and IAAO were 49.52~424.4 U·g^(-1),25.49~84.61 U·(g.min)^(-1),29.77~77.94 U·(g.min)^(-1),198.23~342.94 g FW·min and 40.22~137.2 ug IAA·(h·ml)^(-1),respectively;The highest rooting rate was 81.7%in November,followed by 81.2%in December.Therefore,the cuttings can be taken in November and December under temperature and humidity,in order to obtain the asexual propagated plants of Pinus massoniana.The finding can also provide the theoretical basis for the Masson pine cuttings in production.
