摘要
目的探讨将全基因测序(WGS)技术运用于沙门菌血清型鉴定的可行性。方法对2023年南通市感染性腹泻监测项目中分离鉴定的137株沙门菌进行全基因组测序,基于测序数据利用SeqSero 2在线预测各株菌的血清型,并与血清凝集试验结果比较分析。结果137株沙门菌经血清凝集试验的分型结果为29个血清型,全基因测序后在线预测血清型分为28个。两种分型结果总符合率为97.81%,Kappa值为0.98,其中H抗原单相菌的符合率为100.00%,Kappa值为1.00,而H抗原双相菌的符合率为94.74%,Kappa值为0.94。结论基于WGS数据在线预测的血清型结果与血清凝集试验鉴定的沙门菌血清型结果具有较高的符合率,在实际工作中可以作为沙门菌血清分型的补充手段。
Objective To evaluate the feasibility of whole genome sequencing(WGS)for Salmonella serotype identification.Methods We performed WGS on 137 Salmonella isolates collected from Nantong city's infectious diarrhea surveillance program in 2023.Serotypes were predicted using the SeqSero 2 and compared with results obtained through standard serum agglutination tests.Results 29 serotypes were identified by serum agglutination test among 137 strains of Salmonella,while 28 serotypes were identified using SeqSero 2 based on WGS.The two typing results had a 97.81%overall coincidence rate and a Kappa value of 0.98.Overall,the H antigen monophasic bacteria had a 100.00%coincidence rate and a Kappa value of 1.00,whereas the H antigen biphasic bacteria had a 94.74%coincidence rate and a Kappa value of 0.94.Conclusions The serotype results predicted online based on WGS of Salmonella have a high coincidence rate with the serotype results of Salmonella identified by serum agglutination test.In practical work,it can be used as a supplementary means of Salmonella serotyping.
