摘要
试验旨在克隆雪峰乌骨鸡STIM激活增强子(STIMATE)基因,并对其进行生物信息学及组织表达分析。以雪峰乌骨鸡组织cDNA为模板,克隆STIMATE基因编码区(CDS)并进行生物信息学分析,采用qPCR技术检测STIMATE基因在100日龄雪峰乌骨鸡心脏、肝脏、肺脏、脾脏、肾脏、腺胃、肌胃、胸肌和腿肌9个组织中的表达水平。结果显示,STIMATE基因CDS长度为897 bp,存在1个同义突变,编码298个氨基酸,其氨基酸序列与红原鸡的相似性为100%。STIMATE在雪峰乌骨鸡各个组织中均有表达,其中在肺脏中的表达量最高,在胸肌中的表达量最低。研究表明,试验结果可为进一步探讨STIMATE基因在鸡体内的功能提供参考。
The aim of this study was to clone the STIM-activating enhancer(STIMATE)gene of Xuefeng black-bone chickens and analyze its bioinformatics and tissue expression.The cDNA of tissue was used as template to clone the CDS region of STIMATE gene and sequence it which was analyzed by online software for bioinformatics analysis.qPCR technology was used to detect the expression level of STIMATE gene in nine tissues of heart,liver,lung,spleen,kidney,proventriculus,gizzard,breast muscle and thigh muscle.The results showed that the CDS length of the STIMATE gene was 897 bp,with one synonymous mutation encoding 298 amino acids,and its amino acid sequence was 100%similar to that of Gallus gallus.STIMATE was expressed in various tissues of Xuefeng black-bone chicken,among which the expression level was the highest in the lungs and the lowest in the pectoral muscles.The study indicates that the experimental results can provide a reference for further exploring the function of the STIMATE gene in chickens.
