摘要
试验旨在明确新疆维吾尔自治区和田地区某羊场羊群发病的病原,并特别关注鲍曼不动杆菌作为人兽共患病原菌的可能性。针对患有呼吸道疾病的羊进行了鼻腔拭子样本采集,分离出1株优势菌株。对样本进行细菌分离,经形态学鉴定、生化试验、16S rRNA基因序列对比、药敏试验及小鼠致病性试验等方法对分离株进行综合分析。结果显示,分离株为鲍曼不动杆菌,革兰氏阴性菌,短杆状;生化特性与鲍曼不动杆菌符合;16S rRNA序列与GenBank中鲍曼不动杆菌的同源性为97.2%,命名为HT-1A。药敏试验结果显示,分离株仅对头孢他啶、庆大霉素敏感;致病性试验结果显示,分离株对小鼠具有较强的致病作用,半数致死量(LD50值)为0.6×10^(9) CFU。研究表明,导致羊群发病的致病菌为鲍曼不动杆菌,且该菌株对小鼠具有显著的致病性和致死性。这一结果强调了鲍曼不动杆菌作为人兽共患病原菌,需关注其在动物和人类之间的传播途径。
This study aimed to identify the causative pathogen responsible for the disease outbreak in a sheep flock at a farm in Hotan Prefecture,Xinjiang Uygur Autonomous Region,with particular focus on the zoonotic potential of Acinetobacter baumannii.Nasal swab samples were collected from sheep exhibiting respiratory disease,leading to the isolation of one dominant bacterial strain.The isolate underwent comprehensive analysis through bacterial isolation,morphological identification,biochemical testing,16S rRNA gene sequencing,antimicrobial susceptibility testing,and mouse pathogenicity assays.Results identified the isolate as Acinetobacter baumannii(designated HT-1A),a Gram-negative,short rod-shaped bacterium.Its biochemical characteristics aligned with Acinetobacterbaumannii,and 16S rRNA sequence analysis revealed 97.2%homology with Acinetobacter baumannii strains in GenBank.Antimicrobial susceptibility testing showed the isolate was susceptible only to Ceftazidime and Gentamicin.Pathogenicity testing demonstrated high virulence in mice,with a median lethal dose(LD50)of 0.6×10^(9)CFU.This study confirms Acinetobacter baumannii as the causative agent of the sheep disease outbreak,with the strain exhibiting significant pathogenicity and lethality in mice.These findings underscore Acinetobacterbaumannii's role as a zoonotic pathogen and highlight the need to investigate its transmission routes between animals and humans.
